Cannabinoidi naturali
9-TETRAHYDROCANNABINOL (THC)

In EXPERIMENTAL ANIMALS:
THC induces tolerance and dependence like other drugs of abuse
In MAN:
recent studies indicate that 10-20% of young cannabis users become dependent
Adolescenti
I consumatori di altre droghe

CREB in depressive-like behaviour
In the nucleus accumbens (NAc)
Depressive like behaviour correlated with increase in CREB activity paralleled by increase in dynorphin
levels.
In the prefrontal cortex (PfCtx) and hippocampus (Hippo)
Antidepressant drugs increase pCREB levels

Increased pCREB and dynorphin levels are present in the nucleus accumbens of adult female rats preexposed to THC in adolescence
Conclusion 1• Adolescence exposure to THC induces long term consequences in adult
behavior characterized by sexual dimorphism.
• Female rats exhibited depressive like behavior with anhedonia. This picture was supported by region specific changes in pCREB activation
• Male rats exhibited reduced response to natural rewarding stimuli, in according with previous data of cannabinoiod-induced lacking or blunted responses of Dopamine neurons to pharmacological stimuli (Pistis et al 2004).
• The dysregulation in the CB1 receptor signalling in adult brain areas belonging to the emotional circuit might be a part of the altered emotional responses
Sex-dependent alteration in emotional behavior in adulthood is induced by Cannabis consumption in adolescence.

synaptic plasticity?synaptic plasticity?
Hippocampus
adolescent developmental phase represents a vulnerable time period for persistent effects on synaptic plasticity that could underline adverse actions of
cannabinoids in adulthood. These effects are sex-dependent.
To quote a recent report from the medical branch of the National Academy of Sciences,
“Sex does matter. It matters in ways that we did not expect. Undoubtedly, it matters in ways that we have not yet begun to
imagine.”
