21 monkeys were used to study the analgesic effects of drugs with a warm water tail-withdrawal procedure. These monkeys have their tails immersed in 40-50C water. The tail is not constrained and the monkeys can remove their tails without restriction.

Fifty Two Guinea Pigs Reported Under Reporting Code E:

Guinea pigs are utilized to evaluate the efficacy and toxicity of investigational antifungal agents for the treatment of pulmonary aspergillosis. The advent of sophisticated life saving medical procedures, such as organ transplantation, intensive cytotoxic chemotherapy against cancer, and the prevalence of HIV infection in recent years have dramatically altered the immune status of the patient

I _ - - population during the past ten years. Consequently, the incidence of Aspergillus infection has increased significantly among cancer patients, organ transplant recipients and AIDS patients. Although fungal infection due to A. fumigatus has emerged as a serious clinical problem in immunocompromised patients, the choice of antifungal agents highly effective against A. fumigatus is unfortunately very limited. In this guinea pig model, a variety of novel antifungal agents are evaluated in the treatment of pulmonary aspergillosis. Clinical symptoms of this infection are treated with antifungal agents (except in control animals) and both dietary and fluid supplementation. Animals which do not respond to treatment (and control animals) are euthanized a t the onset of anorexia, fever, dyspnea, or body weight loss greater than 20%.

These sixty-one (61) animals were used in safetyltoxicity evaluations, the results of which will be used to support registration of Pharmacia proprietary compounds. Many of these compounds are being developed for the treatment of critical diseases in humans, primarily central nervous system (CNS) disorders and infectious diseases. Studies to characterize the toxicity of new phamaceuticals are required in non-rodent species by all regulatory agencies that review and approve new human medications, and alternative methodologies alone are not sufficient. Exceptions to Standards and Regulations

The following exceptions to standards and regulations under the Act were approved by the IACUC during this reporting period:

The use of complete Freund's adjuvant (CFA) as part of an immunogen in horses was approved. The immunization process is required in the outline of manufacture for a biological that is the drug of choice for treatment of aplastic anemia and which is also an important part of adjunct therapy in transplant patients. This protocol was not utilized during the current reporting year and therefore impacted zero (0) animals. The IACUC closely monitors this protocol when it is active and is kept current on all efforts

: to refine the procedure by the investigators.

Under one protocol, the IACUC has approved an exemption such that cat enclosu---- ---- ---- --------- ----- ----------- ------- ----- --------- ------ ----------- ------ ----- -------- ----- ----------- --------- ------- ----- ------ --- --------- ------------ -------------- ----------- --- ------------ ------ ------------ ------- -------- . Because of the risk of contaminating other rooms if the cats were moved during sanitization, the rooms were only sanitized between studies (every 6-12 weeks). Granting an exception from the two-week sanitization schedule had little effect on the living conditions of the animals. The rooms and runs were washed down each day. WhiIe the washing proceeds, the cats jump onto perches within the runs and do not get wet. This allows the lower part of the runs and floors to be cleaned daily. During inspections, there have been no concerns about cleanliness. This regimen does allow a small deposit of dust and dander to accumulate on the top rails of the runs, but this is minimal. When a study is completed, runs are washed and disinfected before new animals enter the space.

Under another protocol, the IACUC approved an increase in the sanitization interval for dog runs from 14 days to up to 28 days, total. This impacted 35 dogs during this reporting period. These dogs were on studies where food consumption data was critical, and this was markedly disrupted by moving the animals out of the room in order to clean, adding an additional variable to the study. Granting this exception from the two-week sanitization schedule has had little effect on the living conditions of the animals. The runs are washed down each day (which seems minimally disruptive to the animals' food consumption), and during inspections, there have been no concerns about cleanliness. At the end of each 4-week study, runs are washed and sanitized per usual.

Under two protocols, dogs and nonhuman primates (NHPs) were occasionally housed in stainless steel metabolism cages for the collection of urine andlor feces (providing at least 1 X space) for pharrnacokinetic studies and studies involving radiolabeled compounds, necessitating limited exemptions from the enrichment plans for these species. This impacted thirty-six (36) dogs and twenty- seven (27) NHPs during this reporting period. The dogs were housed in metabolism cages for 1-3 days. They did not have access to manipulata or exercise outside of the cage during this time, due to the need for total urine collection and the use of radioactivity. The NHPs were housed in metabolism cages for 1- 3 days. They did not have access to manipulata throughout this study period, but received fruit treats daily.

Short-term exemptions from exercise for post-surgical recovery were granted by the IACUC as follows: Eight (8) dogs underwent surgical procedures required to prepare them for participation in cardiovascular monitoring studies. These animals were exempted from exercise for a 14-day recovery period, after which time they were able to resume normal activity.

The IACUC has authorized the Attending Veterinarians to grant very short-term exemptions to the mandatory sanitization intervals under exceptional circumstances, providing that they personally view and evaluate the housing situation and determine that there would be no compromise to animal welfare standards by allowing the cage change to wait for one or two additional days. In these situations, the Attending Veterinarian must observe the room in question, then annotate the room maintenance record (RMR), stating that the exception has been granted. A copy is made of the RMR and a separate file documenting these exceptions is maintained in the Attending Veterinarian's office as well. Seven exemptions of this nature were granted this reporting period, each involving a 24-hour extension. Two of these involved a-total of -50 rabbits; three involved a total of 36 dogs; and huo involved a total of 32 nonhuman primates.

Three dogs were euthanized and one died at first dose.)

The animals remained active, alert, and good appetites with no overt signs of pain.

The nature of these test articles required the use of the Maximization method. The FDA requires skin sensitization testing.

Please find enclosed the original and one copy of the completed USDA Annual Report (APHIS form 7023 only) for the reporting facility of The Dow Chemical Company (registration number 34-R-0008, Customer Number 115). This facility has adhered to the standards and regulations under the Animal Welfare Act, as indicated on the Annual Report form.

As note the AHPIS form 7023 has been corrected to reflect the current telephone area code, which is 989, and the correct facility name, whch should be the Toxicology and Environmental Research and Consulting Laboratory. These changes were made on Animal Welfare Registration form submitted in June 2002.

Also enclosed are two copies of the list of former sites, previously listed under USDA . registration number 34-R-0008, which have been permanently closed and are no longer in operation. There are no animal numbers to report from these facilities, as no animals were present in the permanently closed facilities during this reporting period.

The only site that listed animals on this Annual Report is:
