Mucoci l iary t r a n s p o r t is considered t o be an important defense mechanism a g a i n s t i nha led tox ican t s and microorganisms. Never the less , l i t t l e is known about pul­monary c i l i a , t h e i r s t r u c t u r e , how they func t ion , and how t h e i r a c t i v i t y i s con t ro l l ed .

Most knowledge of t h e movement and biochemistry of c i l i a has been der ived by e x t r a p o l a t i o n from work wi th u n i c e l l u l a r organisms o r i n v e r t e b r a t e s . Although the biochemistry and t h e b a s i c mechanisms of m o t i l i t y may be the same i n mam­malian ce i l ia as i n t h e more primitive ones, t h e p a t t e r n of movement and mechanism of c o n t r o l are s p e c i a l i z e d f e a t u r e s t h a t vary from organism t o organism and from t i s s u e t o t i s s u e . No f u n c t i o n a l s t u d i e s of mammalian lung c i l i a can r e l i a b l y be regarded as r e f l e c t i n g t h e normal s i t u a t i o n because the a c t i v i t y of the c i l i a , and the amount and na tu re of s e c r e t o r y a c t i v i t y , may be per turbed by p r e p a r a t i v e procedures .

Recent ly , primary abnorma l i t i e s i n t h e f i n e s t r u c t u r e of human b ronch io la r c i l i a have been observed i n persons wi th s i t u s inve r sus , chronic s i n u s i t i s and b r o n c h i e c t a s i s (Kartagener 's t r i a d ) which i s a n i n h e r i t e d syndrome a s s o c i a t e d wi th repea ted pulmonary i n f e c t i o n s . But whether o t h e r g e n e t i c d e f e c t s i n c i l i a r y s t r u c t u r e may be a s s o c i a t e d wi th pulmonary d i s o r d e r s i s e n t i r e l y unknown. Nor is i t known how exogenous ( t o x i c a n t s , pharmacological agen t s ) o r endogenous (hormones, mediator) compounds a f f e c t c i l i a and t h , e i r func t ion .

The purpose of t h i s announcement is t o encourage submission of r e sea rch g ran t a p p l i c a t i o n s which focus on i n vivo o r i n v i t r o s t u d i e s of t h e s t r u c t u r e and func t ion of normal mammalian c i l i a from t h e upper lower r e s p i r a t o r y t ract . The mechanism of membrane t r a n s f e r of water, e l e c t r o l y t e s , d rugs , and macro-molecules have been s t u d i e d i n t e n s i v e l y over t h e p s s t 20 yea r s i n t h e gastro­i n t e s t i n a l t r a c t , t he s k i n and the r e n a l t ubu le s , and ac ross b loodfb ra in and b lood /b i l e b a r r i e r s , and have l e d t o va luable t h e r a p e u t i c d i scove r i e s . S imi l a r s t u d i e s , however, have not been carr ied o u t i n the lung. A v a r i e t y of ch ron ic and progress ive pulmonary d i s e a s e s r e s u l t from t h e i n h a l a t i o n of o rgan ic o r i no rgan ic material. Agents which are known t o produce c e l l u l a r i n f i l t r a t i o n and/or f i b r o s i s i n the lung inc lude such d i v e r s e subs tances as pigeon droppins , fung i , act inomycetes , n i t r o f u r a n compounds, s i l i c a , and bery l l ium salts. Although s t u d i e s have i n v e s t i g a t e d t h e in f luence of t h e s e agents on lung d i s e a s e , l i t t l e is known regard ing the c e l l u l a r responses of t h e lung t o t h e s e and o t h e r inha led agents o r of t he e x t e n t t o which these responses are p r o t e c t i v e o r d e s t r u c t i v e . Inha led subs tances may produce changes i n immunologicaily-active cells r e s u l t i n g i n de t r imen ta l e f f e c t s which may o r may not be of an immunologic na tu re . It is a l s o p o s s i b l e t h a t a u t o r e a c t i v e responses , such as t h e product ion of a n t i n u c l e a r ant ibody, might arise as a r e s u l t of pre l iminary c e l l u l a r damage caused by a n inha led agen t , and t h a t such auto-immune phenomena might p lay a r o l e i n ch ron ic lung d i s e a s e i n i t i a t e d by t h e i n c i t i n g agent . The purpose of t h i s announcement is t o encourage i n v i t r o s t u d i e s of t h e h o s t response of t he lung t o f i b r o s i s and/or c e l l u l a r - i n f i l t r a t i o n -producing agents .

These sources were e s t a b l i s h e d i n t h e f a c e of c u r r e n t and pending shor t ages i n nonhuman pr imates t o a s s u r e a supply of animals f o r National I n s t i t u t e s of Health (NIH) and Alcohol, Drug Abuse, and Mental Health Adminis t ra t ion (ADAMHA) supported p r o j e c t s . The Diabetes , Endocrinology, and Metabolic Diseases Program of t h e Nat iona l I n s t i t u t e of A r t h r i t i s , Metabolism, and Diges t ive Diseases announces the continued a v a i l a b i l i t y of suppor t f o r manpower development programs i n t h e areas of c l i n i c a l and b a s i c s c i ences r e l evan t t o i t s mission. The programs f a l l i n t o t h r e e c a t e g o r i e s :

.1. Nat iona l Research Serv ice Awards These awards are made t o i n d i v i d u a l s ( fe l lowships) o r t o i n s t i t u t i o n s ( t r a i n i n g programs) f o r t he suppor t of fu l l - t ime research t r a i n i n g r e l a t e d t o d i a b e t e s , endocrinology, and metabol ic d i seases .

2. Research Career Development Award This 'program is employed t o f o s t e r t he development of s c i e n t i s t s wi th outs tanding r e sea rch p o t e n t i a l who r e q u i r e experience i n a product ive s c i e n t i f i c environment i n p repa ra t ion f o r a c a r e e r of independent research . This award provides s a l a r y suppor t f o r f i v e yea r s t o i n d i v i d u a l s who have completed s i g n i f i c a n t p o s t d o c t o r a l research .

3 . Clinical I n v e s t i g a t o r Award This award provides the oppor tuni ty f o r promising c l i n i c a l l y - t r a i n e d i n d i v i d u a l s wi th demonstrated a p t i t u d e i n r e sea rch t o develop i n t o independent i n v e s t i g a t o r s . It provides t h r e e y e a r s of suppor t f o r t he s a l a r y and research ac t iv i t ies of i n d i v i d u a l s wi th four t o seven yea r s of c l i n i c a l and r e sea rch exper ience beyond the c l i n i c a l degree. Like the Research Career Development Award, t h i s award i s designed t o provide exper ience i n a product ive s c i e n t : i f i c environment i n p r e p a r a t i o n f o r a c a r e e r of independent research .

Although t h e DEMD Program i n v i t e s a p p l i c a t i o n from q u a l i f i e d i n d i v i d u a l s f o r each of these awards, t h e Program is p a r t i c u l a r l y i n t e r e s t e d i n encouraging t h e submission of i n s t i t u t i o n a l Nat iona l Research Serv ice Award a p p l i c a t i o n s . This Program permi ts g r e a t e r f l e x i b i l i t y t o t h e p r i n c i p a l i n v e s t i g a t o r i n making commitments and appointments of i n d i v i d u a l s t o the Program, as w e l l as be ing a mechanism e s p e c i a l l y s u i t a b l e f o r exposing the t r a i n e e s o r fe l lows t o a v a r i e t y of t r a i n i n g exper iences .
