Vevo770 –Probe head (707B/704) Preparation

•  The acoustic membrane is very fragile and sensitive, it must be handled with care.

•  The probe head should be filled with water all the time. If it has been drained; follow the procedures on this page.

–  Position the RMV scan head with the fill port pointing upward.

–  Fill the nosepiece with de-ionized water using the supplied water bottle until the nosepiece is almost full.

–  Using your thumb, cover the fill port and tap the scanhead several times to dislodge any air bubbles in the nosepiece.

–  Check to ensure that the face of the transducer does not contain bubbles. If bubbles appear, tilt the RMV scanhead to dislodge them.

–  Orient the nosepiece so that the remaining air bubbles are placed under the fill port, and finish filling the nosepiece with the syringe.

–  Insert and tighten the fill port screw with a screwdriver. Do not over-tighten the fill port.

Pre-Image Preparation-Animal Preparation

•  Warm up the stage –  The stage warmer, heart and temperature monitors each have a simple on/off button on the control pad.

•  Located on the table to the left of the stage •  Turn on the stage heater-this helps to maintain mouse

body temperature

•  Study Subject Preparation –  ultrasound is most effective with no hair.

•  Depilate the scan area of the animal using a razor or depilatory cream (Nair) •  Be sure to remove the NAIR as it can easily burn the mouse

–  Put electrode contact gel (Sigma) or imaging gel on all 4 of the foot pads on stage base. Tape mouse down on platform so that all four of its feet are on the appropriate foot pad and in contact with the probe.

•  This will monitor the mouse’s heart rate and respiration •  the heart rate should be between approximately 370 and 400 beats per minute

–  Prior to placing imaging gel on the mouse, take a small amount of gel and smooth it over the surface of the image area to remove small air bubbles at the skin level.

–  Place 1-2 inches of gel on the image area, lower the scan head into the gel. Make sure to avoid bubbles.

Set up a study •  Turn on the instrument and computer

–  The system main power switch is a large toggle switch on the back right hand side of the image.r

–  PC power is activated by small toggle switch on the left side of the unit near the DVD drive-If this switch is not working, the main computer power button is below the console.

–  Each user will log on to VEVO770 using your Kerberos username and password. You will be billed for the greater of your reserved time or your logged time. Please have the same person make reservations and log on/off of the instruments.

•  Open the Visual Sonics program, from the desktop icon –  If an error code comes up simply click okay.

•  Initialize scan head •  Select or add operator •  Set up new study, click the New tab •  Fill in spaces for user and project info •  Select scanning mode from the keypad

–  B-mode: basic scanning, two dimensional images and cine loops.

–  Doppler Mode: measures velocity of blood flow from a field of view (FOV).

–  Power Doppler: measures flow volume or noise in 2 or 3-D from an FOV.

Image acquisition/saving

•  For all modes, start imaging in B-mode to identify an area of interest.

–  If you want to have respiration and cardiac information included in the session, press the view tab on the tools menu.

–  On the left side of the screen uncheck box for cardiac mode, with cardiac mode on, images will vibrate.

–  Select the desired features.

•  To save a single image select “Frame Store”, press “Ctrl +L” to label and save data.

Menu tab View

•  To save a video loop up up to 300 frames press “Scan Freeze”; this tab will both start and stop data acquisition. The last 300 frames will be saved, to save the data press “Cine Store” then “Ctrl +L’’ to label and save the movie. •  At the conclusion of your study, in the Tool menu Study tab Close Study; Commit Session Data tab to save.

Data transfer to the CCRBI2 server

•  Data Transfer to the server -To transfer your data to the ccrbi2 file server you

will need to map the drive as follows: • Computer>tools>map network drive •  In the folder line type the following:

\\ccrbi2\ccrbi2_Data1\atwai\ (there is an underscore between ccrbi2 and Data1, not just a space)

• You will be asked for a username and password •  User name: ccrbi2\malstrom •  Password: Ray4eiph

B-Mode: basic scanning, two dimensional images and cine loops

The line in this icon corresponds to the orientation ridge on the RMV scanhead and indicates the orientation of the RMV scanhead relative to the image.

B-Mode ultrasound image window.

To toggle between the basic and detailed display parameters views, click Details or press <Ctrl+D>.

The grayscale bar indicates the dynamic range of the display.

Cine loop playback controls.

Physiological Data trace.

The cine loop range control slider defines a subset of the cine loop to be played back or saved.

The depth ruler indicates the depth from the transducer face. The triangular arrow indicates the focal length of the transducer.

The time-gain compensation (TGC) sliders specify the receive gain to be used for acquiring different sections of a B-Mode image.

Define a Field of View (FOV) for PW Doppler studies

•  Defining the Sample Volume and Doppler angle must be done in B-Mode.

•  Defining the PW Doppler Sample Volume. –  Select Overlay > PW Doppler Sample Volume Wire-frame –  Move the trackball cursor over the red vertical line, left up

click to move sample volume –  The trackball cursor is displayed as a crosshair when it is

within a region that can be repositioned.

–  Left-click the position line. Use the trackball cursor to move the overlay.

–  Left-click again to place the Sample Volume. The Sample Volume details are updated in the parameters display.

•  Defining the PW Doppler Angle. –  Start acquiring B-Mode data. –  From the B-Mode window, position the trackball cursor over

the –  PW Doppler angle line (displayed as a dotted yellow or blue

line). The trackball cursor changes to a crosshair.

–  Right up-click to activate editing of the PW Doppler angle line. The angle line follows the movement of the trackball cursor.

–  When the angle is at the desired setting, right-click to set the angle.

–  VisualSonics recommends a PW Doppler angle that is between 0° and 60° for the most accurate velocity estimate.
