Der ACE Drugcup Enterprise Drogentest-Kit ist ein Schnelltest für den simul-tanen und qualitativen Nachweis von Drogen und Drogenmetaboliten im menschlichen Urin. Mit diesem Test können folgende Drogen nachgewiesen werden: Amphetamin, Kokain, Ecstasy, Marihuana, Methamphetamin und Opiate / Morphin.

VERWENDUNGSZWECKDer Urine Cup ist ein chromatographischer Immunotest für den qualitativen Nachweis von Drogen und Drogenmetaboliten im menschlichen Urin. Der Test weist Kombinationen der nachfolgend aufgelisteten Drogen in den jeweils angegebenen Konzentrationen nach.

TESTPRINZIP Der Urine Cup ist ein kompetitiver, chromatographischer Immunotest. Mögliche in der Urinprobe vorhandene Drogen konkurrieren mit der jeweiligen chemisch markierten Droge (Drogen-Protein-Konjugat) um Bindungsstellen an dem spezi-fischen anti-Drogen-Antikörper.

Bei der Messung wandert eine Urinprobe auf Grund der Kapillarwirkung durch die Testmembran. Eine vorhandene Droge unterhalb der Konzentration des Cut-offs blockiert nicht die Bindungsstellen des spezifischen Antikörpers. Der spezifische Antikörper reagiert mit dem Drogen-Protein-Konjugat und eine farbige, sichtbare Testlinie zeigt sich. Beim Vorhandensein von Drogen oberhalb der Cut-off Konzentration werden alle Bindungsstellen des anti-Dro-gen-Antikörpers von der Droge blockiert und es kann keine farbige Linie an der Testlinie ausgebildet werden.

Folglich bedeutet eine Linie im Testbereich ein negatives Ergebnis für die Droge, während das Fehlen einer Testlinie im Testbereich ein positives Ergebnis für die Droge anzeigt.

Eine farbige Linie wird immer im Bereich der Kontrolllinie erscheinen und dient damit als Verfahrenskontrolle, die das korrekt zugefügte Probenvolumen und Membrandurchfeuchtung bestätigt.

LAGERUNG UND STABILITÄTDer Urine Cup kann versiegelt in der Schutzfolie, bei Raumtemperatur oder gekühlt (2 - 30°C) bis zu dem auf der Folie aufgedruckten Ablaufdatum gelagert werden. Das Produkt ist feuchtigkeits-empfindlich und sollte direkt nach dem Öffnen der Schutzfolie verwendet werden. Kits sollten vor dem Sonnenlicht geschützt aufbewahrt werden. Š Nicht einfrieren Š Nicht nach dem angegebenen Haltbarkeitsdatum verwenden.

INTERPRETATION DER ERGEBNISSE

Negativ*: Zwei Linien erscheinen. Eine Linie sollte im Kontrollbereich (C) liegen, die andere direkt darunter im angrenzenden Testbereich (T). Das negative Ergebnis besagt, dass die Drogenkonzentration unter der Nachweisgrenze liegt. Positiv: Nur eine farbige Linie erscheint im Kontrollbereich (C). Im Testbereich (T) erscheint keine Linie. Dieses positive Ergebnis besagt, dass die Drogen-konzentration über der Nachweisgrenze liegt.Ungültig: Es erscheint keine Kontrolllinie. Ungenügende Probemenge und falsches Vorgehen sind die häufigsten Gründe für das Fehlen einer Kontroll-linie. Das Vorgehen prüfen und den Test mit einer neuen Testvorrichtung wiederholen. Besteht das Problem weiter, nutzen Sie diese Testcharge nicht mehr weiter und nehmen Sie Kontakt mit Ihrer Bezugsquelle auf.*HINWEIS: Der Farbton im Testbereich (T) kann unterschiedlich sein. Sobald aber selbst eine schwache farbige Linie erkennbar ist, muss dies als negatives Ergebnis gewertet werden.

TESTDURCHFÜHRUNG1. Bringen Sie vor dem Testbeginn die Urinprobe und / oder Kontrollen auf Raumtemperatur (15 - 30°C). 2. Bringen Sie den Beutel vor dem Öffnen auf Raumtemperatur. 3. Nehmen Sie den Test aus der Schutzfolie und verwenden Sie ihn so bald wie möglich.4. Die Urinprobe wird direkt in den Cup gegeben und der Deckel fest zugeschraubt, sodass kein Probenmaterial auslaufen kann.5. Stoppen Sie die Zeit und warten Sie darauf, dass die roten Linien bei den Drogenteststreifen erscheinen.6. Beschriften Sie das Label mit den Patientendaten.7. Überprüfen Sie den Temperatur-Teststreifen 2-4 Minuten nach der Proben-abgabe. Eine grünliche Verfärbung zeigt an, ob die Temperatur der Probe zwischen 32°C und 38°C liegt. Eine frische unverfälschte Urinprobe sollte eine Temperatur von 32-38°C haben.

QUALITÄTSKONTROLLEAls testinterne Kontrolle sind im Bereich der Kontrolllinie C spezifische Antikörper vorhanden, die mit überschüssigem Reagenz eine rote Linie bilden. Die Ausbildung dieser C-Linie zeigt an, dass ausreichendes Probenvolumen aufgetragen wurde, die Chromatographie einwandfrei abgelaufen ist und die Antigen-Antikörper-Komplexbildung nicht gestört ist.

Eine externe Kontrolle wird nicht mit diesem Test mitgeliefert. Es wird im Rahmen der “Guten Laborpraxis” empfohlen, dass positiv und negativ Kontrollen getestet werden, um die Testdurchführung und die korrekte Testleistung zu überprüfen.

EINSCHRÄNKUNGEN1. Dieser Test liefert ausschließlich ein vorläufiges, qualitatives analytisches Ergebnis. Eine zweite analytische Methode muss zur Bestätigung der Resultate eingesetzt werden. Gaschromatographie / Massenspektrometrie (GC / MS) oder Flüssigkeits-Chromatographie / Massenspektrometrie (LC / MS) stellen die bevorzugten Methoden dar.2. Es besteht die Möglichkeit, dass technische oder verfahrensbedingte Fehler, ebenso wie störende Substanzen in der Urinprobe zu fehlerhaften Ergebnissen führen.3. Verfälschungsmittel wie Bleichmittel und / oder Alaun können in Urinproben fehlerhafte Ergebnisse, unabhängig von der verwendeten analytischen Methode, erzeugen. Wenn eine Verfälschung vermutet wird, sollte der Test mit einer weiteren Urinprobe und einem neuen Cup wiederholt werden.4. Ein positives Ergebnis zeigt das Vorhandensein der Droge oder ihrer Metaboliten an, jedoch nicht das Ausmaß der Intoxikation, die Art der Verabrei-chung oder die Konzentration im Urin.5. Ein negatives Ergebnis bedeutet nicht unbedingt, dass die Probe keine Drogen enthält. In der Probe können sich dennoch Drogen in einer Konzen-tration unterhalb der Nachweisgrenze des Assays befinden.6. Dieser Test unterscheidet nicht zwischen Medikamenten- / Drogenmiss-brauch oder einer anderen Einnahme von Medikamenten.

REAGENZIEN Der Test besteht aus einem Membranstreifen, auf dem sich eine Testlinie aus Drogen-Rinderprotein Antigenkonjugat und eine Kontrolllinie, aus Ziege- anti-Kaninchen-Antikörper, befindet und einem Farbstoff-Pad, das monoklonale Antikörper und Kaninchen Antikörper Goldkomplex enthält.

Analytische Sensitivität Die Sensitivität des Urine Cup Tests wurde durch GC / MS bestätigte Kontrollen bei Konzentrationen von negativ, -50% Cut-off, -25% Cut-off, Cut-off, +25% Cut-off, +50% Cut-off und 3 X Cut-off bestimmt. Die Ergebnisse sind nachfolgend zusammengefasst:

KreuzreaktivitätEine Studie wurde durchgeführt, um die Kreuzreaktivität des Tests zu bestimmen. Dafür wurde ein drogenfreier PBS Puffer mit verschiedenen Substanzen versetzt. Die folgenden Verbindungen führten bei Testungen mit dem Urine Cup Test bei Konzentrationen von bis zu 100 g / mL nicht zu falsch-positiven Resultaten.

EINSCHRÄNKUNGENEs ist nicht möglich alle auf dem Markt befindliche Medikamente, außer die in der Gebrauchsanweisung aufgeführten, auf eine Kreuzreaktion oder einen anderen Einfluss auf den Test zu prüfen.

Wenn der Patient einen „Cocktail“ aus mehreren, unterschiedlichen Medika-menten einnimmt, kann nicht ausgeschlossen werden, dass eine nicht mehr nachvollziehbare Kreuzreaktion das Testergebnis verfälscht.

The ACE Drug Cup Enterprise Drug Test Kit is a rapid test for the simultaneous and qualitative detection of drugs and drug metabolites in human urine. With this test the following drugs can be detected: Amphetamine, Cocaine, Ecstasy, Marijuana, Methamphetamine and Opiates / Morphine.

SAMPLING AND STORAGE Š The Urine Cup is intended for human urine only. Š The urine sample can be collected at any time. Š The urine sample must be collected in a clean, dry cup. Š Turbid urine specimens should be centrifuged, filtered or stored so that

turbidity can settle and a clear urine specimen is available. Š Urine samples may be stored at 2 - 8°C for up to 48 hours. Š For long-term storage, the urine specimen should be frozen at -20°C.

Frozen urine specimens should be thawed, brought to room temperature and mixed thoroughly before testing.

TEST ROUTINE The Urine Cup is a competitive chromatographic immunoassay. Possible drugs present in the urine sample compete with the respective chemically labelled drug (drug-protein conjugate) for binding sites to the specific anti-drug antibody.

During the measurement, a urine sample migrates through the test membrane due to the capillary effect. An existing drug below the cut-off concentration does not block the binding sites of the specific antibody. The specific antibody reacts with the drug-protein conjugate and a colored, visible test line appears.

In the presence of drugs above the cut-off concentration, all binding sites of the anti-drug antibody are blocked by the drug and no colored line can be formed on the test line. Consequently, a line in the test area indicates a negative result for the drug, while the absence of a test line in the test area indicates a positive result for the drug.

A coloured line will always appear in the area of the control line and serves as a procedural control confirming the correctly added sample volume and membrane moisture penetration.

STORAGE AND STABILITYThe Urine Cup can be stored sealed in the protective pouch, at room temperature or refrigerated (2-30°C) until the expiry date printed on the pouch. The product is sensitive to moisture and should be used immediately after opening the protective film. Kits should be stored away from sunlight. Š Do not freeze. Š Do not use after the specified expiration date.

INTERPRETATION OF RESULTS

Negative Positive Invalid

Negative*: Two lines appear. One line should be in the control area (C), the other directly below in the adjacent test area (T). The negative result indicates that the drug concentration is below the detection limit. Positive: Only one colored line appears in the control area (C). No line appears in the test area (T). This positive result indicates that the drug concentration is above the detection limit.Invalid: No control line appears. Insufficient sample quantity and incorrect procedure are the most common reasons for the absence of a control line. Check the procedure and repeat the test with a new test device. If the problem persists, stop using this test batch and contact your source.* NOTE: The color in the test area (T) can be different. However, as soon as even a weak colored line is visible, this must be interpreted as a negative result.

DIRECTIONS FOR USE1. Bring the urine sample and / or controls to room temperature (15-30°C) before starting the test.2. Bring the pouch to room temperature before opening. 3. Take the test out of the pouch and use it as soon as possible.4. The urine specimen is placed directly into the cup and the lid is tightened so that no specimen material can leak.5. Stop the time and wait for the red lines to appear on the drug test strips.6. Label the label with the patient data.7. Check the temperature test strip 2-4 minutes after sample delivery. A greenish discoloration indicates whether the temperature of the sample is between 32°C and 38°C. A fresh unadulterated urine specimen should have a temperature of 32-38°C.

QUALITY CONTROLSpecific antibodies are present in the area of control line C as an internal test control, which form a red line with excess reagent. The formation of this C-line indicates that sufficient sample volume has been applied, chromatography has been performed correctly and antigen-antibody complex formation has not been impaired.

An external control is not supplied with this test. It is recommended in Good Laboratory Practice that positive and negative controls are tested to verify test performance and correct test performance.

LIMITATIONS1. This test provides only a preliminary qualitative analytical result. A second analytical method must be used to obtain a confirmed result. Gas chromatography / mass spectrometry (GC / MS) or liquid chromatography / mass spectrometry (LC / MS) are the preferred methods.2. There is a possibility that technical or procedural errors, as well as other interfering substances in the urine specimen may cause erroneous results. 3. Adulterants, such as bleach and / or alum, may produce erroneous results in urine samples, regardless of the analytical method used. If adulteration is suspected, the test should be repeated with another urine specimen and a new cup.4. A positive result indicates the presence of the drug or its metabolites, but not the extent of intoxication, method of administration or concentration in the urine.5. A negative result does not necessarily mean that the sample does not contain drugs. However, drugs may be present in the sample at concentrations below the detection limit of the assay.6. This test does not distinguish between drug of abuse or any other medication.

REAGENTS The test consists of a membrane strip containing a test line of bovine drug protein antigen conjugate and a control line of goat anti-rabbit antibodies, and a dye pad containing monoclonal antibodies and rabbit antibody gold complex.

CROSS-REACTIVITYA study was conducted to determine the cross-reactivity of the test. A drug- free PBS buffer was mixed with various substances. The following compounds did not lead to false positive results when tested with the Urine Cup test at concentrations of up to 100 g / mL.

RESTRICTIONSIt is not possible to test all drugs on the market, except those listed in the instruction manual, for a cross-reaction or other effect on the test. If the patient is taking a “cocktail” of several different drugs, it cannot be ruled out that a cross-reaction that can no longer be traced will falsify the test result.
