Accumulation of genetic mutations causes normal types of cells to transform to cancerous types.
Cancer driver mutations, such as in EGFR, TP53, KRAS, BRAF, BRCA1/2, and MET, play a
significant role in tumor development. These essential mutant genes have become therapeutic
targets for a number of first-line targeted drugs in cancer treatment. However, drug resistance
and limited extending of survival time are the bottlenecks of targeted drug treatment.
Therefore, the development of new approaches and finding new targets are required.
One of the representative successful targeted drugs is the EGFR-TKI (tyrosine kinase inhibitor) in
lung cancer treatment1. Four EGFR-TKIs (erlotinib, gefitinib, afatinib and osimertinib) are
available for the treatment of patients with EGFR positive mutations. Although tumor cells with
mutant EGFR are extremely sensitive to EGFR-TKIs, acquired resistance is inevitable2. Therefore,
there is an imperative to understand resistant mechanisms and find new therapies.
CRISPR/Cas9 is a powerful tool for elucidation of resistant mechanisms, identification of new
targets, as well as a potential cancer therapy3. In this thesis, we aimed to explore the potential
of CRISPR/Cas9 for discovering therapeutic targets as well as for cancer therapy. To this end, we
first reviewed the applications of CRISPR/Cas9 in the identification of new cancer targets, drug
resistance and cancer therapy. Later, we used CRISPR/Cas9 to knockout EGFR from different
cancer cells, investigated the alterations of other Her family receptors, downstream signaling
and the interactions of other receptors, including CXCR7, vEGFR and PDGFR. Despite the rapid
development of CRISPR/Cas9 technique, the gene editing potential of CRISPR/Cas9 is not yet
sufficient enough for cancer therapy. Therefore, we sought to improve the CRISPR/Cas9 gene
editing efficiency by altering the chromatin state through modulation of the HDAC and HAT
activity.
In Chapter 2, we comprehensively reviewed and summarized the applications of CRISPR/Cas9
for uncovering new cancer targets and therapy. We provide an overview of studies in which
CRISPR/Cas9 has been utilized for the identification of potential therapeutic targets in some of
the most frequent solid tumors, including lung, breast, brain, liver, and colorectal cancer. We
discuss potential candidates for therapy that are either highly expressed or activated in different

potential therapeutic targets, such as CD38, CXCR2, MASTL, and RBX2, as well as several
noncoding (nc) RNAs have been identified using CRISPR/Cas9 technology4.
In Chapter 3, we summarized the importance of CRISPR/Cas9 system in understanding drug
resistance mechanisms and identification of resistance-related genes such as PBRM1, SLFN11
and ATPE1 in different cancers. We also provided an overview of genes, including RSF1, CDK5,
and SGOL1, whose disruption can synergize and/or re-sensitize the currently available drugs
such as paclitaxel and sorafenib3.
Tyrosine kinase inhibitors (TKIs), such as erlotinib, gefitinib, and afatinib, are the commonly used
targeted agents for NSCLC patients with certain EGFR mutations5. However, inevitably, almost
all treated patients treated with TKIs develop resistance within 9-13 months after treatment
initiation5,6. Mechanisms of EGFR-TKI resistance in NSCLC are complicated. In order to deepen
our understanding of EGFR targeting resistance, in Chapter 4, we focus on characterizing of
EGFR ablation in NSCLC cells using CRISPR/Cas9 and further investigating other HER family
receptors behaviors upon EGFR loss or resistance. We observed distinct and surprising
behaviors when cells loss of mutant EGFR compared to the loss of wild type EGFR, including cell
proliferation, migration and downstream signaling pathways. Our findings demonstrate that
HER2 and HER3 overexpression and transcriptional activation of cyclin D1 is a mechanism of TKI
acquired resistance in NSCLC.
KRAS-driven non-small cell lung cancer (NSCLC) patients have no effective targeted treatment1.
In Chapter 5, we investigated EGFR knockout as a therapeutic option in EGFR wild type and
KRAS mutated lung cancer cells7. To the best of our knowledge, this is the first study showing
that wildtype EGFR plays a significant role in the growth of KRAS-dependent tumor cells. We
identified the overexpression of CXCR7 as a bypass mechanism in EGFR-/- cells by promoting
MAPK signaling. Both EGFR and CXCR7 inhibition showed synergetic suppression of tumor cell
growth. Furthermore, we revealed that CXCR7 was increased in EGFRwt, but decreased in
EGFRmut cell lines after treatment with gefitinib. We also show that CXCR7 expression is higher

in adenocarcinoma (ADC) than squamous cell lung carcinoma (SQCC) in NSCLC patients and the
healthy lung tissue. Therefore, dual inhibition of EGFR and CXCR7 might be a potential
treatment strategy for patients with NSCLC.
Since EGFR plays a significant role in various cancers, we focus on EGFR targeting and expand
our study from lung cancer to renal cancer8,9. In Chapter 6, we examined whether EGFR
knockout in combination with different small molecular inhibitors or therapeutic proteins (TRAIL)
can be used as a therapeutic approach for RCC. We showed that disruption of overexpressed
EGFR dramatically inhibits the proliferation of RCC and arrests cells at G2/M checkpoint.
Furthermore, we found that inhibition of PDGFR and VEGFR by sunitinib can attenuate the
expression of pERK1/2 and pAKT induced by EGFR loss. Thus, ablation of overexpressed EGFR by
CRISPR/Cas9 or in combination with sunitinib may be an option for renal cell carcinoma
treatment.
Despite the rapid development of CRISPR/Cas9-mediated gene editing technology, the gene
editing potential of CRISPR/Cas9 is hampered by low efficiency, especially for cancer gene
therapy10,11. In Chapter 7, we comprehensively investigated the impact of HDACs and HATs on
CRISPR/Cas9 mediated gene editing. We found that Entinostat (HDAC1/2/3 inhibitor) and
Panobinostat (pan-HDAC inhibitor) enhanced Cas9 gene editing activity while other inhibitors
(HDAC4/5/6/7/8/9) did not. We also found that RGFP966 (HDAC3 inhibitor) decreased
CRISPR/Cas9 gene editing activity, which can be used for downregulation CRISPR/Cas9 gene
editing activity. Importantly, Entinostat and Panobinostat dramatically increase gene knock-in
rates. We confirmed these findings by knockdown of HDAC1, HDAC2 and HDAC3. We further
identified that HDAC inhibition (Entinostat and Panobinostat) facilitated Cas9 access to target
DNA and increased cutting frequencies. Our study revealed an essential role of HDACs in
CRISPR/Cas9 mediated gene editing. We demonstrate that it is feasible to modulate
CRISPR/Cas9 gene editing efficiency by regulating HDAC activity. This method might also be
widely used in regulating other nucleases (ZFNs, TALENs, CRISPR/Cas9 and dCas9)-mediated
genome and epigenome editing.

Future perspectives
We aimed to use CRISPR/Cas9 for cancer research and enhancing CRISPR/Cas9 for cancer
therapy in this thesis. We concentrate on EGFR targeting by CRISPR/Cas9 and compared to
EGFR-TKIs treatment. We uncovered three compensating or alternative mechanisms in EGFR
targeting: HER2/3 overexpression and transcriptional activation of cyclin D1 (Chapter 4), CXCR7
overexpression (Chapter 5), vEGFR and PDGFR activating (Chapter 6).
Although EGFR ablation by CRISPR/Cas9 can mimic EGFR–TKIs treatment in some aspects,
disruption of EGFR by CRISPR/Cas9 is different from TKI treatment. Since EGFR-TKI may only
inhibit the tyrosine kinase activity of EGFR, other functions of EGFR may remain active or being
more active12. However, EGFR knocked out by CRISPR/Cas9 will remove all of EGFR functions
and may lead to activation of alternative pathways7,13. Future studies need to elucidate the
differences and correlations between knockout and inhibition. To this end, investigation of
transcriptome and proteome levels alteration upon loss of EGFR may give more insights.
Specifically, in Chapter 5, our data showed that EGF can still stimulate pAkt in EGFR-/- cells. This
observation indicates that EGF-induced phosphorylation of Akt may be independent of EGFR,
which has not been reported previously to our knowledge. One hypothesis is that EGF
stimulates pAkt via CXCR7, because previous studies have shown that CXCR7 associated
pathways can induce Akt phosphorylation14,15 and we observed CXCR7 overexpression in A549
EGFR-/- cells7. In addition, one recent study reported that Akt activation is related to EGFR drug
resistance in lung cancer16. Of note, a study reported that MIF O-GlcNAcylated MIF binds to
EGFR17. Other studies show that MIF is a ligand of CXCR715,18. Collectively, it is rational to
speculate whether EGF or DDT (MIF2) is a ligand of CXCR7. Given the importance of these
interactions, future studies are needed to explore the crosstalk between EGFR, CXCR7 and their
ligands (EGF, DDT, MIF and others) (Figure 1)

We have set up a platform for screening the small molecular compounds for enhancing or
inhibiting CRISPR/Cas9 activity (Chapter 7). Using this platform, we provide a practical option for
improving gene editing efficiency through chromatin de-condensation using HDAC inhibition.
Furthermore, our study facilitates a deeper understanding of gene editing process by altering
the accessibility of the DNA through histone acetylation and histone deacetylation. This
platform can be widely used for screening enhancers and inhibitors of nucleases mediated-gene
editing for future studies (Figure 2).

Future perspectives
We aimed to use CRISPR/Cas9 for cancer research and enhancing CRISPR/Cas9 for cancer
therapy in this thesis. We concentrate on EGFR targeting by CRISPR/Cas9 and compared to

Opeenstapeling van genetische mutaties zorgen ervoor dat normale cellen zich transformeren
naar kanker cellen. Kanker gerelateerde mutaties, zoals in EGFR, TP53, KRAS, BRAF, BRCA1/ 2 en
MET, spelen een belangrijke rol bij tumorontwikkeling. Deze essentiële gemuteerde genen zijn
therapeutische doelen geworden voor een aantal eerstelijns gerichte geneesmiddelen bij de
behandeling van kanker. Echter, geneesmiddelenresistentie en beperkte verlenging van de
overleving zijn de knelpunten van gerichte medicamenteuze behandeling. Daarom zijn de
ontwikkeling van nieuwe behandelingen en het vinden van nieuwe aangrijpingspunten vereist.
Een van de belangrijkste succesvolle gerichte medicijnen is de EGFR-TKI (tyrosine kinase remmer)
bij de behandeling van longkanker. Vier EGFR-TKIs (erlotinib, gefitinib, afatinib en osimertinib)
zijn beschikbaar voor de behandeling van patiënten met EGFR-positieve mutaties. Hoewel
tumorcellen met een gemuteerde EGFR extreem gevoelig zijn voor EGFR-TKIs, is verworven
resistentie onvermijdelijk. Daarom is het noodzakelijk om de resistentiemechanismen te
begrijpen en om nieuwe therapieën te vinden.
CRISPR/Cas9 is een krachtig hulpmiddel voor het ophelderen van resistentiemechanismen,
identificatie van nieuwe aangrijpingspunten en is zelf een potentiële therapie tegen kanker. In
dit proefschrift onderzochten we de toepassing van CRISPR/Cas9 bij de ontdekking van nieuwe
therapeutische doelen, waaronder nieuwe therapieën bij de behandeling van kanker. We
hebben eerst de toepassingen van CRISPR/Cas9 beoordeeld bij de identificatie van nieuwe
aangrijpingspunten, resistentiemechanismen en kankertherapie. Later gebruikten we
CRISPR/Cas9 om het EGFR gen uit te schakelen in verschillende kankercellijnen. Ook
onderzochten we de veranderingen in andere receptoren van de Her-familie, signaaltransductie
en de interactie met andere receptoren zoals CXCR7, vEGFR en PDGFR. Ondanks de snelle
ontwikkeling van de CRISPR/Cas9 techniek is zijn mogelijkheid om veranderingen in genen aan
te brengen nog niet efficiënt genoeg voor kanker therapie. Daarom zochten we een manier om
de efficiëntie van CRISPR/Cas9 te veranderen. We deden dit door de staat van het chromatine

te variëren via aanpassingen in de activiteit van bepaalde enzymen, de zogenaamde HATs en
HDACs
In hoofdstuk 2 worden de toepassingen van CRISPR/Cas9 bij de ontdekking van nieuwe
aangrijpingspunten voor de behandeling van kanker uitvoerig besproken. We geven een
overzicht van studies waarbij CRISPR/Cas9 ingezet werd voor de identificatie van potentieel
nieuwe therapeutische doelen in enkele van de meest voorkomende solide tumoren zoals long-,
borst-, hersen-, lever- en colorectaal kanker. We bespreken potentiele kandidaten voor therapie.
Omdat deze kandidaten hoog tot expressie komen of zeer actief zijn in diverse kankersoorten,
zijn ze het meest eenvoudig te remmen. We laten zien dat diverse therapeutische targets, zoals
CD38, CXCR2, MASTL en RBX2 als ook een aantal non-coding (nc) RNAs zijn geïdentificeerd met
behulp van de CRISPR/Cas9 technologie4.
In hoofdstuk 3 wordt de relevantie van het CRISPR/Cas9 systeem met betrekking tot het
begrijpen van geneesmiddelresistentie en de identificatie van resistentie-gerelateerde genen
zoals PBRM1, SLFN11 en ATPE1 in diverse soorten kanker samengevat. Ook geven we een
overzicht van onder andere de genen RSF1, CDK5 en SGOL1, waarbij de verstoring hiervan
tumoren opnieuw gevoelig kan maken voor onder andere de al beschikbare geneesmiddelen
paclitaxel en sorafenib3.
Tyrosine kinase remmers (TKIs) zoals erlotinib, gefitinib en afatinib zijn de meest gebruikte en
gerichte middelen bij patiënten met NSCLC met bepaalde mutaties in EGFR5. Het is echter
onvermijdelijk dat bijna alle patiënten die met TKIs behandeld worden resistentie ontwikkelen
binnen 9-13 maanden na de start van de behandeling5,6. De mechanismen achter EGFR-TKI
resistentie in NSCLC zijn zeer complex. Om meer te weten te komen over EGFR gerichte
resistentie focussen we ons in hoofdstuk 4 op het karakteriseren van NSCLC cellen waarbij EGFR
is uitgeschakeld met behulp van CRISPR/Cas9. Ook onderzoeken we het gedrag van andere
receptoren die behoren bij de Her-familie na EGFR uitschakeling of resistentie. We hebben
duidelijk en verrassend gedrag waargenomen bij cellen waarbij het gemuteerde EGFR
uitgeschakeld werd ten op zichte van cellen waarbij wild type EGFR werd uitgeschakeld. We

zagen een verschil in o.a. cel proliferatie, cel migratie en in downstream signaalroutes. Onze
bevindingen laten zien dat HER2 en HER3 overexpressie en de transcriptionele activiteit van
cyclin D1 een mechanisme is van TKI verworven resistentie in NSCLC.
Voor patiënten met NSCLC met KRAS-gerelateerde niet-kleincellige longkanker bestaat nog geen
effectief gerichte behandeling1. In hoofdstuk 5 hebben we een EGFR knock-out onderzocht als
mogelijke therapie. We deden dit in zowel een wild type als in KRAS-gemuteerde longkanker
cellen7. Voor zover wij weten is dit de eerste studie waarin aangetoond werd dat wild type
EGFR een belangrijke rol speelt in de groei van KRAS-afhankelijke tumor cellen. We lieten zien
dat de overexpressie van CXCR7 een bypass mechanisme is in EGFR-/- cellen door het stimuleren
van de MAPK signalering. Zowel EGFR als CXCR7 remming lieten een synergetische
onderdrukking van de tumorgroei zien. Verder hebben we aangetoond dat de hoeveelheid
CXCR7 was toegenomen in EGFRwt, maar juist was afgenomen in EGFRmut cellijnen na
behandeling met gefitinib. We lieten ook zien dat de expressie van CXCR7 hoger is in NSCLC
patiënten met adenocarcinoma (ADC) vergeleken met plaveiselcelcarcinoom (SQCC) en gezond
longweefsel. Daarom kan een dubbele remming van zowel EGFR als CXCR7 een mogelijke
behandelingsstrategie zijn voor patiënten met NSCLC.
Omdat EGFR een significante rol speelt in verschillende typen tumoren, focussen we ons op de
targeting van EGFR maar breiden we ons studie uit van longkanker naar niercelkanker8,9 (RCC).
In hoofdstuk 6 onderzoeken we of een EGFR knock-out in combinatie met diverse kleine
moleculaire remmers of therapeutische eiwitten (TRAIL) gebruikt kunnen worden als een
therapeutische benadering voor RCC. We lieten zien dat een verstoring van tot overexpressie
gebrachte EGFR de proliferatie van cellen dramatisch remt en ze laat stilstaan op het G2/M
checkpoint tijdens de celdeling. Verder vonden we dat de remming van de PDGFR en VEGFR
door sunitinib de expressie van pERK1/2 en pAKT, geïnduceerd door het verlies van EGFR, kan
afzwakken. Oftewel, ablatie van een tot over expressie gebrachte EGFR door CRISPR/Cas9 al dan
niet gecombineerd met sunitinib kan een optie zijn voor de behandeling van niercelkanker.

Ondanks de snelle ontwikkeling van de CRISPR/Cas9-gemedieerde gen-editing technologie
wordt zijn potentieel verminderd door een lage efficiëntie, in het bijzonder bij kanker
gentherapie10,11. In hoofdstuk 7 hebben we de invloed van HDACs en HATs op CRISPR/Cas9
gemedieerde gen-editing uitgebreid onderzocht. We vonden dat Entinostat (een HDAC1/2/3
remmer) en Panobinostat (pan-HDAC remmer) de activiteit van CRISPR-Cas9 gen-editing
verhoogden terwijl andere remmers (van HDAC4/5/6/7/8/9) dat niet deden. Ook vonden we dat
RGFP966 (specifieke HDAC3 remmer) de gen-editing activiteit van CRISPR/Cas9 verminderde,
welke daarom gebruikt zou kunnen worden voor de downregulatie van CRISPR/Cas9. Belangrijk
is dat Entinostat en Panobinostat de knock-in frequentie verhoogden. Dit werd bevestigd door
de knock-out van HDAC1, HDAC2 en HDAC3. Daarnaast ontdekten we dat de remming van
HDACs (door Entinostat en Panobinostat) de toegang van Cas9 tot het target DNA faciliteert en
daar het aantal breuken in het DNA verhoogt. Onze studie laat de essentiële rol van HDACs in
CRISPR/Cas9 gemedieerde gen-editing zien. We tonen aan dat het haalbaar is om de efficiëntie
van CRISPR/Cas9 te moduleren door middel van het veranderen van de HDAC activiteit. Deze
methode zou ook op grote schaal toegepast kunnen worden voor het reguleren van andere
nuclease (ZFNs, TALENs, CRISPR/Cas9 en dCas9) gemedieerde genoom en epigenoom
bewerkingen.
Toekomstperspectieven
In dit proefschrift wilden we de CRISPR/Cas9 technologie gebruiken voor onderzoek naar kanker
en wilden we de activiteit van CRISPR/Cas9 verbeteren voor kankertherapie. We focusten ons
op de EGFR targeting door CRISPR/Cas9 en vergeleken dit met behandeling met EGFR-TKi. We
ontdekten drie compenserende of alternatieve mechanismen in EGFR targeting: HER2/3
overexpressie en verhoogde transcriptionele activiteit van Cyclin D1 (Hoofdstuk 4), CXCR7
overexpressie (Hoofdstuk 5) en vEGFR en PDGFR activatie (Hoofdstuk 6).
Hoewel de ablatie van EGFR door CRISPR/Cas9 de behandeling met EGFR-TKIs in sommige
aspecten kan nabootsen verschillen deze twee van elkaar. Aangezien EGFR-TKIs alleen de
tyrosine kinase activiteit van EGFR remmen, blijven de overige functies van EGFR actief of

worden zelfs actiever12. Hoewel een knock-out van EGFR door CRISPR/Cas9 alle functies van
EGFR verwijderd, kan het leiden tot de activering van alternatieve routes7,13. Toekomstige
studies zijn nodig om het verschil en de correlatie tussen knock-out en remming op te helderen.
Nieuw onderzoek naar verandering van transcriptoom- en proteoomniveaus na verlies van EGFR
moet hierin meer inzicht geven.
In hoofdstuk 5 toonde onze data specifiek aan dat EGF nog steeds pAkt kan stimuleren in EGFR-
/- cellen. Deze waarneming geeft aan dat EGF-geinduceerde fosforylatie van Akt mogelijk
afhankelijk is van EGFR, hetgeen zover ons bekend nog niet eerder is gerapporteerd. Een
hypothese is dat EGF pAKT stimuleert via CXCR7, aangezien andere studies hebben laten zien
dat CXCR7 geassocieerde routes pAKT fosforylatie kunnen induceren14,15 en omdat wij hebben
waargenomen dat CXCR7 tot overexpressie wordt gebracht in EGFR-/- A549 cellen7. Bovendien
heeft een recente studie aangetoond dat de activatie van Akt gerelateerd is aan de EGFR
gemedieerde geneesmiddelenresistentie bij longkanker16. Opgemerkt moet worden dat een
studie rapporteerde dat MIF O-GlcNAcylated MIF aan EGFR bindt17. Andere onderzoeken tonen
aan dat MIF een ligand is van CXCR715,18. Alles bij elkaar is het redelijk te speculeren dat EGF of
DDT (MIF2) een ligand is voor CXCR7. Gezien het belang van deze interacties is er meer
onderzoek nodig om de “cross-talk” tussen EGFR, CXCR7 en zijn liganden (EGF, DDT , MIF en
anderen) te onderzoeken (Figuur 1).

Ondanks de snelle ontwikkeling van de CRISPR/Cas9-gemedieerde gen-editing technologie
wordt zijn potentieel verminderd door een lage efficiëntie, in het bijzonder bij kanker
gentherapie10,11. In hoofdstuk 7 hebben we de invloed van HDACs en HATs op CRISPR/Cas9
gemedieerde gen-editing uitgebreid onderzocht. We vonden dat Entinostat (een HDAC1/2/3
remmer) en Panobinostat (pan-HDAC remmer) de activiteit van CRISPR-Cas9 gen-editing
verhoogden terwijl andere remmers (van HDAC4/5/6/7/8/9) dat niet deden. Ook vonden we dat
RGFP966 (specifieke HDAC3 remmer) de gen-editing activiteit van CRISPR/Cas9 verminderde,
welke daarom gebruikt zou kunnen worden voor de downregulatie van CRISPR/Cas9. Belangrijk
is dat Entinostat en Panobinostat de knock-in frequentie verhoogden. Dit werd bevestigd door
de knock-out van HDAC1, HDAC2 en HDAC3. Daarnaast ontdekten we dat de remming van
HDACs (door Entinostat en Panobinostat) de toegang van Cas9 tot het target DNA faciliteert en
daar het aantal breuken in het DNA verhoogt. Onze studie laat de essentiële rol van HDACs in
CRISPR/Cas9 gemedieerde gen-editing zien. We tonen aan dat het haalbaar is om de efficiëntie
van CRISPR/Cas9 te moduleren door middel van het veranderen van de HDAC activiteit. Deze
methode zou ook op grote schaal toegepast kunnen worden voor het reguleren van andere
nuclease (ZFNs, TALENs, CRISPR/Cas9 en dCas9) gemedieerde genoom en epigenoom
bewerkingen.
Toekomstperspectieven
In dit proefschrift wilden we de CRISPR/Cas9 technologie gebruiken voor onderzoek naar kanker
en wilden we de activiteit van CRISPR/Cas9 verbeteren voor kankertherapie. We focusten ons
op de EGFR targeting door CRISPR/Cas9 en vergeleken dit met behandeling met EGFR-TKi. We
ontdekten drie compenserende of alternatieve mechanismen in EGFR targeting: HER2/3
overexpressie en verhoogde transcriptionele activiteit van Cyclin D1 (Hoofdstuk 4), CXCR7
overexpressie (Hoofdstuk 5) en vEGFR en PDGFR activatie (Hoofdstuk 6).
Hoewel de ablatie van EGFR door CRISPR/Cas9 de behandeling met EGFR-TKIs in sommige
aspecten kan nabootsen verschillen deze twee van elkaar. Aangezien EGFR-TKIs alleen de
tyrosine kinase activiteit van EGFR remmen, blijven de overige functies van EGFR actief of

worden zelfs actiever12. Hoewel een knock-out van EGFR door CRISPR/Cas9 alle functies van
EGFR verwijderd, kan het leiden tot de activering van alternatieve routes7,13. Toekomstige
studies zijn nodig om het verschil en de correlatie tussen knock-out en remming op te helderen.
Nieuw onderzoek naar verandering van transcriptoom- en proteoomniveaus na verlies van EGFR
moet hierin meer inzicht geven.
In hoofdstuk 5 toonde onze data specifiek aan dat EGF nog steeds pAkt kan stimuleren in EGFR-
/- cellen. Deze waarneming geeft aan dat EGF-geinduceerde fosforylatie van Akt mogelijk
afhankelijk is van EGFR, hetgeen zover ons bekend nog niet eerder is gerapporteerd. Een
hypothese is dat EGF pAKT stimuleert via CXCR7, aangezien andere studies hebben laten zien
dat CXCR7 geassocieerde routes pAKT fosforylatie kunnen induceren14,15 en omdat wij hebben
waargenomen dat CXCR7 tot overexpressie wordt gebracht in EGFR-/- A549 cellen7. Bovendien
heeft een recente studie aangetoond dat de activatie van Akt gerelateerd is aan de EGFR
gemedieerde geneesmiddelenresistentie bij longkanker16. Opgemerkt moet worden dat een
studie rapporteerde dat MIF O-GlcNAcylated MIF aan EGFR bindt17. Andere onderzoeken tonen
aan dat MIF een ligand is van CXCR715,18. Alles bij elkaar is het redelijk te speculeren dat EGF of
DDT (MIF2) een ligand is voor CXCR7. Gezien het belang van deze interacties is er meer
onderzoek nodig om de “cross-talk” tussen EGFR, CXCR7 en zijn liganden (EGF, DDT , MIF en
anderen) te onderzoeken (Figuur 1).

We hebben een platform opgezet voor de screening van kleine moleculaire verbindingen die de
activiteit van CRISPR/Cas9 verhogen of verlagen (Hoofdstuk 7). Met behulp van dit platform
bieden we een praktische oplossing voor het verhogen van de gen-editing efficiëntie via de de-
condensatie van het chromatine door middel van HDAC remming. Bovendien geeft onze studie
een beter begrip van het proces van gen-editing door de toegankelijkheid van het DNA met
behulp van histon acetylatie en histon deacetylatie te veranderen. Dit platform kan voor vervolg
studies op grote schaal worden gebruikt voor de screening van versterkers en remmers van
nuclease gemedieerde gen-editing (Figuur 2).
