INTRODUCTION
Potato production in Indonesia continue to decrease, due to use of late generation potato tubers as seeds
Use of late generation has caused high disease infestation, particularly virus
Need to use high quality, early generation seed potatoes
Department of Agriculture, Bali and a number of farmers in Bedugul region, has started to use early generation seed potatoes.
Need to confirm the quality of seed
Research aim:
• To find out virus symptoms on early generation (G1) at Kembang Merta Village
• To confirm virus type using DAS-ELISA technique
Research Method
Symptoms observation
Horticulture Field Station, Kembang Merta Village, Tabanan, Bali
Sample were taken from G1 potato field at 3 different screen house
Fresh leaves with virus symptom were taken and keep on cool box and brought to lab
DAS – ELISA METHOD
Plate filled 100 μl antibody +
wrapped using tissue &
aluminum foil.
Overnight
Weigh each sample 4 grams & crushed
until smooth
Plate washed 3x with PBST
The scoured liquid is inserted
into the plate (100 μl)
according to the elite & incubation map ± 2 hours
Plate washed 8 x with PBST
Enter each 100 μl conjugate at each sample & incubation well
± 2 hours
Plate washed 8 x with PBST Enter PNPP (100 μl) in
each well
Incubate room temperature
30-60 minutes
ELISA reader
2. Serological test (DAS-ELISA) results
Plants were virus positive: PVX, PVS, PVY and PLRV at all three potato screen house.
Conclusion and suggestion
Conclusion :
1. Virus symptoms observed were: mosaic, leaf roll, rugose, malformation and mottle.
2. Early generation seed potato were infected with PVX (81,81 %), PVS (27,27 %), PVY (27,27 %) and , PLRV (18,75 %).
Sugestion :
1. Improve screen house isolation and pest control should be done more intensively
