The existing magnet system, stand and helium transfer line will be integrated with new NMR components.We are replacing digital electronics, RF amplifiers, and the cryoplatform as well as adding NMR probes and sample changing capabilities. This instrument will be available to UF researchers through the AMRIS facility. The instrument will provide state of the art capabilities for structural characterization of natural products, of interest for drug development; measurements of biomolecular structure and dynamics, for functionally characterizing proteins and nucleic acids; and metabolomics measurements, enabling quantitative determination of changes in key metabolites of importance to disease diagnoses. Optimum sensitivity is important because only limited sample quantities are available. Interleaved experiments enable significant time savings. The samples studied on these instruments require the highest sensitivity and reliability available in terms of the digital electronics, amplifiers and NMR probes utilized. In particular, cryogenically cooled electronics enable NMR cryoprobes to achieve 2- to 4-fold gains in sensitivity over conventional NMR probes at the same magnetic field. Cryoprobes operating at 14.1 are only available from a single vendor, Bruker Biospin, and have the highest sensitivity specifications available at these magnetic fields. Bruker only sells directly to customers. They are the sole vendor for NMR systems incorporating cryogenically cooled NMR cyroprobes and multiple receivers for measurements at magnetic fields of 14.1 T and above. There are no other vendors that could supply this equipment. A second consideration in studying challenging samples is the ability to interleave NMR experiments characterizing different NMR nuclei. The new NEO architecture offered by Bruker is the only NMR platform available with inherent multi-receive architecture that enables simultaneous characterization of signals from hydrogen, carbon, and nitrogen nuclei enabling full characterization of fragile biological samples in a fraction of the time required using conventional NMR instruments. This will enable structural characterization of natural products, of interest for drug development; measurements of biomolecular structure and dynamics, for functionally characterizing proteins and nucleic acids; and metabolomics measurements, enabling quantitative determination of changes in key metabolites of importance to disease diagnoses. A third consideration in determining our instrument configuration is the dynamic range of the radiofrequency receivers. For samples containing complex mixtures of metabolites, a broad dynamic range for the receiver enables the detection of molecules over a broad range of concentrations, as is typically present in biological fluids. Bruker Biospin currently offers the best receiver options in terms of dynamic range.
