Use SGT’s LC/MS filter on yourNitrogen line to deliver 99.9999% puregas. Flow is split within a 2-positionbaseplate so that half gets sent toeach high surface area charcoal trap,and then is rejoined at the exit of thebaseplate (see Figure 1). These car-tridge-style filters maintain the “fast-connect” principle which means they“snap” onto the self-sealing basepla-te. Spectrum provides new technologydeveloped by the National CancerInstitute for screening of anti-cancercompounds. This in vivo method ofidentifying compounds with potentialchemotherapeutic activity againstcancer and HIV is now routinely usedby many researchers. The technologyallows human cells to be implantedinto host animals and subsequentlyretrieved in mass. While in the host,cells can be exposed to potentialchemotherapeutic compounds asantiviral agents and the impact of that

exposure can be measured afterretrieval. With this technology, targetcells are macroencapsulated in bio-compatible hollow fibers. Thesemacroencapsulated cells are thenimplanted into the subcutaneous orintraperitoneal compartments of lab-oratory mice. This methodology is asignificant step forward and results insignificant savings of time, labor,amount of compound required, andthe number of animals used in theevaluation of the potential therapeuticactivity of candidate compounds.

The hollow fiber assay is a unique invivo model permitting the simultane-ous evaluation of compound efficacyagainst six cell lines at subcutaneous(s.c.) and peritoneal (i.p.) sites(Hollingshead et al. Life Sci. 57;131, 1995). This model was used toinvestigate the relationship betweencell density and compound activity.Additionally, the model was adaptedfor mechanism of action studies. Thistechnology is now used routinely bythe National Cancer Institute of theNIH for testing compounds for anti-cancer activity. It is also beingemployed to look for antiviral com-pounds for HIV.

The membranes, made from modifiedPolyvinylidene Difluoride (mPVDF), repre-sent a revolutionary advance in separationtechnology. The membranes are biocom-patible, hydrophobic and resistant to awide variety of organic solvents, as wellas, most aqueous acids and bases. Themembranes can be heat sealed and auto-claved without affecting the molecularweigh cut off (MWCO) or changing themembrane performance.

CellMax Implant Membranes, the uniquepigmented hollow fiber with the ability toheat seal, is ideal for controlled release,cell culture, encapsulation and implanta-tion research. The MWCO is 500,000Daltons.

The surface of the CellMax Implant

Membranes has been shown to be biocompatible in various animal modelsfor periods exceeding 14  days.Multiple cell lines have been grown with-in the CellMax Implant Membranes bothin vitro and in vivo. Cell lines grown withinCellMax Implant Membranes, which havebeen implanted into a host animal, are notsubject to immunological attack by thehost animal. CellMax ImplantMembranes containing cancer cell lineshave been implanted into mice and usedto screen for compounds having anti can-cer activity. Similarly, CellMax ImplantMembranes containing HIV infected celllines have been implanted into mice andused to screen for compounds havinganti-HIV activity. A Kit For the Optimization of ProteinPurfication Strartegy

Purification of a novel protein is often adifficult undertaking. If a researchlaboratory is not sufficiently experien-ced in dealing with the purification ofnovel proteins then the task of purify-ing a protein could become a very fru-strating undertaking. Considerabletime and money may simply be wastedin testing various purification strate-gies.

The Protein Master- STRATEGYTM is akit designed to simplify the protein

purification and optimization procedu-res. The STRATEGYTM uses carefullyplanned protein extraction buffers,methods of protein estimation, andstep-by-step purification protocols.When carefully followed, protein purifi-cation can be optimized within a fewdays.

The kit guides the researcher step-by-step through various protein purifica-tion techniques, which include variousprotein precipitation-fractionation andchromatography techniques. TheProtein Master- STRATEGYTM is supp-lied with agents needed to test various

purification techniques. No expensiveequipment is needed.

Our mono-component, liq-uid TMB solutions combine TMB, sub-strate buffer and hydrogen peroxide fora safe, ready-to-use, one step, chro-mogen system. The products do notcontain harmful solvents.Recommended for ELISA procedures,liquid TMB yields a soluble blue endproduct read at 655 nm. The TMB Blotting/TMB Blotting PLUSare ready-to-use substrates containing3,3´, 5, 5´ tetramethylbenzidine andhydrogen peroxide. These substratesare intended for use in blotting proce-

dures with peroxidase labelled anti-bodies. The positive reaction gives astable blue colour, and the productsgive a good sensitivity and a low back-ground staining. Stability life time is 21/2 years at 4°C.

OPD, Ortho-Phenylenediamine, is usedas a chromogen substrate in ELISAprocedures with an absorption maxi-mum at 492 nm and produces a yellowcolour during the enzymatic degrada-tion of hydrogen peroxide by horserad-ish peroxidase. After addition of sulfu-ric acid you will obtain a very stable endsolution. The fizzing, buffer containingtablets are dissolved in distilled waterwithin few minutes. You get the maxi-mum convenience,- an easy, safe andreliable way to operate. Stability lifetime is 18 months at 4°C.

AEC (3-amino-9-ethylcarbazole) isused as a chromogen substrate inimmunoblotting and immunohisto-chemistry. AEC produces a rose-redcolour during the enzymatic degrada-tion of H2SO4 by horseradish peroxi-dase. Each tablet contains 5 mg of AECand dissolves in approx. 5 minutes. 1tablet is added 1 ml of 96 % alcohol,followed by addition of distilled water to10 ml.This will give a solution of pH 5.1+ 0.1 buffered with 20 mM citrate.Stability life time is 18 months at 4°C.

DAB (3,3-diaminobenzidine tetrahy-drochloride), is used as a chromogensubstrate in immunohistochemistry.

DAB produces a brown colour duringthe enzymatic degradation of H2SO4 byhorseradish peroxidase. Each tabletcontains 10 mg of DAB and dissolves indistilled water in less than 5 minutes.Stability life time is 18 months at 4°C.

Fast Red is used as a chromogen sub-strate in immunoblotting and immuno-histochemistry using alkaline phos-phatase antibody conjugates. Eachtablet contains 0.5 mg naphthol phos-phate, 2 mg FAST RED chromogen and0.4 mg levamisole, an inhibitor ofendogenous enzyme activity. Stabilitylife time is 18 months at 4°C.

The Urea Peroxide tablets is the safeand easy alternative for the addition ofhydrogen peroxide. One tablet dis-solved in 10 ml of the substrate solu-tion will give a concentration of 6 mMH2O2. Stability life time is 18 months at4°C.

Avidin is purified from hen's egg whiteby affinity chromatography. The purifi-cation procedure ensures minimal con-tamination by other proteins or DNA.The resulting high activity and purity ofthe product gives very low non-specificbinding.

The HRP is purified by repeated affinitychromatography of root extracts fromhorseradish. The result is an enzyme ofhigh specific activity. Our horseradishperoxidase is purified in large scale bythe use of benzhydroxamic acid

The liquid BCIP/NBT is the most com-monly used substrate for colorimetricdetermination of Alkaline Phosphatase(AP) activity in membrane tests. Weoffer an aqueous buffered solution con-taining NBT (NitroBlue Tetrazolium)and BCIP (Bromo-Chloro-IndolylPhosphate) forming a blue/purple pre-cipitate at the positive sites. The colourdevelops when AP catalyses thedephosphorylation of BCIP. The intenseblue/purple precipitate is very stablealso resisting fading when exposed tolight.

The 5 Minute Reversible Copper StainTM

is a single step stain for the rapiddetection of proteins fractionated bySDS-PAGE. No de-staining is neces-sary. The stain is based on the interac-tion of copper ions with polyacrylamideand proteins. The stain works bydepositing a copper metal precipitatein the gel which turns the gel opaquegreen, while the SDS coating on theproteins prevents the stains from bind-ing to the proteins. A negative image is

produced; clear protein bands aredetected against a semi-opaque bluepolyacrylamide background. Proteinbands are visualized in as little as 5minutes. The sensitivity of the 5 MinuteReversible StainTM is 8-12ng . Stainingdoes not interfere with the electroelu-tion of proteins or alter their biologicalproperties. Gels stained with the 5Minute Reversible Copper StainTM canbe de-stained in 20-25 minutes beforethe transfer or electroelution of pro-teins.
