Title: Production of Keratinolytic Enzyme by A New Isolated Feather Degrading Bacillus sp. From chick feather waste.Author: Mathusamy Govarthanan,. T. Selvankumar, S. ArunprakashSynopsis made by : Ika Dwiyulita, H31114302A novel feather-degrading Bacillus Sp. was isolated from chick manure. The bacterium produces keratinolytic enzyme using chicken feather as a sole of carbon and nitrogen source. The effect of temperature, pH on keratinase production of this keratinolytic strain was studied. In addition, the Bacillus sp was able to completely degrade within a period of 48 hours. Keratinase is an extra cellular enzyme used for the bio degradation of keratin. Keratinase is produced only in the presence of keratin substrate. Bacterial strains are known which are capable of degrading feathers because of their selectivity in degrade the beta keratin present in feathers wich is mean that enzyme in these bacterial having keratinolytic activity.In industry, we get a fact that there is a great demand for developing biotechnological alternatives for recycling of keratin waste by converting the unused chick feather. The curre study focused on isolation and characterization of extracellular keratinase producing Bacillus sp. from chick waste featherThe following steps will guide us to find the result. First, we isolate the kerationlytic micoorganizm by flooded in saline solution, then screening on skim milk agar plates for testing the caseinolytic activity of the organism and the next step is subculturing. Then we identified the morphological, physicological and biochemical of the bacterial by chromogenicmethod. Then we assayed the keratinase activity and also the proteolic activity by using azo-caesin and then we can determine the protein according to the method described by Bradord. We also determine the bacterial growth by total plate count on nutrient agar and calculated the percentage of feather degradation from the differences in residual feather dry weight between a control and treated sample.The newly isolated Bacillus sp. is a novel strain which can degrade feather keratin. The isolated bacterium showed high keratinolytic activity when cultured on feather medium. The optimum temperature of keratinases ranging from 30 to 80C. Bacillus strains are ubiquitous microorganisms, which can grow on natural media without any special requirements. This novel keratinolytic isolate could be a potential candidate for degradation and utilization of feather keratin.
