DIRECTOR'S COLUMN ... P oultry production in India continues to grow at healthier rate (6-8% in layer and 10- 12% in broiler) and provides much needed animal protein at affordable price to common man. However, this also generates sustained demand for agricultural products such as maize, soybean, sunflower meals, rice bran, etc. leading to higher prices of these agricultural commodities, and therefore, the profit margin of poultry farming is under stress. We need to explore alternative or non-conventional feed resources to bring down the cost of feeding in poultry. In a study conducted at DPR, it was demonstrated that alternate feed resource such as acidulated soapstock, a by-product of vegetable oil refining industry has been incorporated by replacing the sunflower oil as the energy source with no adverse effects on the performance of Krishibro broilers. It indicates that this by-product could be used to reduce the expenditure on feed in Krishibro broiler farming. The protocol for gene knockdown of myostatin gene has been perfected to enhance body weight in chicken. Backyard poultry is an important component of low input poultry farming in the country for rural and tribal farmers. The performance of PD-4 (Improved Aseel) birds with respect to egg production up to 72 weeks of age was impressive under intensive type of management. Scientists demonstrated the activity of Matrix metalloproteinases in chicken seminal plasma. Public Private Partnership was emphasised as a way forward for research and development field. DPR has entered in to as many as four memorandums of understanding with private firms for providing consultancy and research services during the period. Notable are the value addition to alternative feed resources and diagnostic services for Infectious Bronchitis virus. DPR stall continues to attract visitors at various exhibitions conducted in the period. Director joins all the staff to wish a Happy New Year - 2015 for readers of the ICAR-DPR News letter. RESEARCH HIGHLIGHTS

Technique to knock-down myostatin gene inchicken perfectedA total of 5 shRNA molecules located at 194, 21S, 29S, SII and926 nucleotide down-stream of the initiation codon of myostatin(MSTN) gene with GC% varied from 47 to 52% were designedand cloned in pENTR U6 entry vector. The shRNA moleculeswere tested in vitro in chicken primary myoblast culture fortheir relative efficiency and found that all the molecules hadsilencing efficiency up to 60% for expression of MSTN gene. Allthe shRNA molecules were transferred through embryo as wellas spermatozoa using DMSO, High temperature (HT) semendiluent and PBS by gene pulser and lipofectamine mediatedmethod. The effect of these molecules on post-hatch bodyweight was analyzed and found Significantly (P<0.05) higherbody weight at 4, 6 and S weeks of age. The molecule 5 showedIS.S, 37.S, 25 and 33 % higher body weight than the birds ofcontrol group at 2, 4, 5 and 6 weeks of age respectively. At 6weeks, molecules 3, 1,2 and 4 showed 26.9, 22, 23.4 and 26.4 %higher body weight than the birds of control group, respectively. The comparison of different methods revealed that gene pulserwith HT diluent for transfection to spermatozoa showedsignificantly highest body weight at 6 weeks of age. This methodwas followed by DMSO simple incubation, gene puls er withPBS and lipofection. This method of knocking down the genemay be used for silencing other deleterious genes to improveproductivity and health of chicken.

Acidulated soapstock as an energy source in thediet of broiler chickensFats/oils are most valuable source of energy in poultry diet.Since oils are expensive, there is continuous search for alternateenergy sources in order to reduce the cost of production. Onesuch energy source is acidulated soapstock (AS), which is aby-product of vegetable oil refining industry and is essentiallya good source of energy and essential fatty acids. The presentstudy was therefore conducted to evaluate the effect ofacidulated soapstock as an energy source in the diet of broilerchicken. Day old broiler (Krishibro) chicks were wing bandedand randomly distributed into 40 raised-floored stainless steelbattery brooder pens with 5 birds per pen. Basal starter (0-3week, 2900 kcal ME/kg and 22% CP) and finisher (4-6 week,

3000 kcal ME/kg and 20% CP) diets were prepared usingsunflower oil (SFO) as an energy source. Subsequently, 3 testdiets were prepared in which SFO was replaced with acidulatedsoapstock at 25, 50 and 100% level. Each diet was fed at randomto 10 replicates of 5 chicks each throughout the experimentalperiod (0-6 week). The body weight and feed consumptionparameters were recorded on weekly basis. At the end of theexperimental period, 10 birds from each dietary group wereslaughtered to record the carcass traits. Dietary replacement ofSFO with AS partially (25 or 50%) or completely (100%) hasno influence on either body weight gain or feed conversionratio. The carcass characteristics such as dressed weight, breastmeat yield, giblet weight and abdominal fat contents were alsonot influenced by the replacement of SFO with AS in the diet. From the results of the present study, it can be concluded thatacidulated soapstock can be used as an energy source in thediet of Krishibro broiler chickens.

Production performance of PD-4 birds up to72 weeks of ageProduction performance of PD-4 birds (Improved Aseel)was evaluated up to 72 weeks of age. It was observed thatsurvivors' egg production up to 72 weeks of age was 156.5±2.79eggs while hen day (HDEP) and hen housed egg production(HHEP) were 144.9 and 137.5 eggs respectively. Survivors' eggproduction recorded up to 40 weeks of age was 60.S± 1.32. Eggproduction started at 21 weeks and peaked during 33-36 weeksand thereafter it declined slowly up to 65-6S weeks but againincreased marginally during 69-72 weeks of age. Egg weightconsistently improved from 35.13 g at age at first egg to 4S.07g at 40 weeks of age. However, it has increased by 1.56 g from40 weeks of age to 72 weeks of age (49.63±0.43). Body weightand shank length of hens at 72 weeks of age were 1949±19.05g and 104.5±0.31 mm respectively. There was moderateimprovement in body weight from 40 weeks of age to 72 weeksof age (1949±19.1).

Effect of organic chromium on breeder semenquality, fertility and hatchability in PD-3lineChromium (Cr), a naturally occurring element in trivalentform has been shown to play important role in glucose, fatand protein metabolism. Supplemental Cr has been reportedto improve semen quality, fertility and hatchability in otherspecies. A feeding experiment was carried out to evaluate theeffects of supplemental organic Cr on semen quality, fertilityand hatchability in PD-3line. A total of 40 PD-3 roosters of 22weeks age were equally distributed in to four groups and dietswere supplemented with organic Cr at graded levels (0, 0.3, 0.6,0.9 mg/kg) for 12 weeks duration. Semen collected after 4 and8 weeks of experiment was analysed for different gross semenparameters and seminal plasma lipid peroxidation. A fertilitytrial was conducted for 4 weeks duration where fixed dose ofsperm (lOOmillion in O.lml) was inseminated every week. Theeggs collected were incubated and fertility and hatchabilitydata was collected. The results indicated no significant effectof supplemental Cr on different parameters studied. It can beinferred that Cr supplementation in diet does not improvebreeder semen quality, fertility and hatchability in PD-3 line.
