Objective- To solve the problem of chitosan/TPP nanoparticles with high degree of monodispersity & stability.
Introduction
Why Monodisperse system?
Why low molecular weight chitosan is is taken as a? polymer
Why ion gelation technique is used for the formation?
4
Chitosan
It is a linear polysaccharide composed of randomly distributed ß-(1-4)- linked D- glucosamine and N-Acetyl D-glucosamine.

Materials and Methods
Mixing FILTER
Filter Filter
LMW Chitosan +Acetic acid solution
Stirred with magnetic stirrer for overnight
PH adjusted to4.7-4.8
Pass through syringe filter
TPP + ULTRA PURE WATER
Pass through syringe filter(0.22µm)
Addition of the both solution under the specific Conditions

Results and discussions
1.Effect of the concentration on both solution-
Size of the particles get increased with increase in concentration.
under acidic condition there is electrostatic repulsion b/w chitosan molecules.
Found that the chitosan under the low con. Could form
stable nanoparticles even at a low mass ratio, while could form only stable nanoparticles at higher conc.
8
2. Effect of mass ratio-
Fig: Effect of the mass ratio of chitosan to TPP on particle size And zeta potential by adding different volumes of TPP to 10ml of Chitosan solution
9
3. Effect of PH of chitosan solution
The effect of the pH of chitosan solution on the formation of chitosan/TPP particles was investigated by adjusting the pH from 3.6 to 5.5 (3.6, 4.0, 4.2, 4.5, 4.7, 5.0, 5.2, 5.5).
The results indicated that the critical mass ratio of chitosan to TPP for the formation of an opalescent suspension decreased with the pH decreasing.
10
4. Effect of Temperature of chitosan solution
Fig.- Effect of temperature of solution on particle size.
11
5. Effect of the ambient temperature during cross- linking
12
6. Effect of the concentration of acetic acid
13
7. Effect of the Stirring speed
The particle size distribution was significantly narrowed by increasing stirring speed from 200 rpm to 800 rpm.
8. Colloidal StabilityEvaluated by storage at room temperature (10–20◦C) for different
periods of time.
The results showed that after stored at room temperature for 20 days, the liquid was still a clear suspension with light blue opalescence and the nanoparticles did not show any statistically significant change in particle size and its distribution during the investigation.
14
Inference- Decreased toxicity
Compatibility with drugs
Good carrier for delivery of drugs
Increased sustained release action
No problem with solubility
Easy to form by this method
