1. Operation Principle

Generally, unprocessed microbial samples are mushy, farinose or

block-like. To facilitate analyzing and inoculating samples, samples shall

be refined to be liquid-like. Basic principle of operation: original samples

(bigger ones shall be cut to be block-like. Dimension: 10×10mm) shall be

put into homogenize bag together with certain liquid or solvent.

Homogenizer will hammer sample bag to generate pressure. The pressure

will arouse surge and accelerate mixing, so microbial components in the

solvent can be equally distributed. This homogenizer can be used to

homogenize brain, kidney, liver, spleen etc.

Homogenize bag made from special materials and used to store

original samples and solvent has gone through asepsis. Seal the bag

with the rail on the top of the homogenizer door can ensure no

leakage of liquid while raped by relevant plate and balanced mixing

of the sample in the bag by the pressure from the rapping. Generally,

the homogenize speed range is 3～～～～12 T/Sec and the time is 0.1～～～～

99min, however, customers can adjust them to fit actual situations.

180 sec and 8 T/Sec are acceptable to the homogenizing of livers of

mouse and rat, for instance.

I. Application and Scope

It is widely used in the asepsis of animal tissues, biological samples

and food (such as pig stomach, meat, fish, vegetables, fruits and

biscuit), and cosmetics. Thanks to its easy operation, convenience and

low time-consuming, it is especially qualified to be used in the

preparation for test samples of various microbes. As single-use

sterilized homogenize bag is soft, pollution-free & damage-free, with

no heating-up, no need to carry out asepsis and wash vessels,

containing glass windows for easy observation, and adopting

qualified and environmental stainless steel or similar materials, it can

avoid cross contamination and protect the operators. Besides, by

homogenizing various tissues of tumor, such as liver cancer, intestinal

cancer, gastric cancer and breast cancer, etc, abundant individual

cells can be obtained at the speed of 2×105

in every 2 min, as well as

adjusting the process of softening these tissue cells.

III. Operation Procedures and Setting

Open the packing case, and carry out the homogenizer, then put

it on a stable working table near power source.

1. Plug in (AC220V, 50Hz) first, then power on.

2. Start the equipment, the boot screen will appear.

3. Standby Screen (Press key)

Indication of the Temperature Control:

1. Green: The set temperature has been

reached.

2. Red: Heating

(1) Modify the timing and rev (for method, refer to aforesaid).

(2) Sterilization (can only be started by pressing key on the

standby screen).

The automotive sterilization period is 5min.

Press key to stop the sterilization process.

(3) The temperature displayed is the actual temperature inside the

trunk and heat preservation begins when the temperature set is

reached.

(4) Press key to the transfer screen of data stored.

4. Short press key to the standby screen, and press it again

to work. Remember: Long press is not effective.

(1) Start on, the indicator light is on ( )

(2) Adjust the rev (rap speed) at real-time.

(3) Short press to pause ( indicator light

flashes), press again to return.

(4) If you long press during running, the equipment will

stop work and back to the standby screen.

5. Check whether the under edge of the homogenizer door has been

buckled to the rail at the bottom of homogenize trunk. Remember,

the under edge of the homogenizer door should be buckled perfectly

to the rail.

6. Hold the door switch and open the door fully with your right hand,

then, put the homogenize bag with samples and solvent into the

homogenize trunk with your left hand. The under edge of the

homogenize bag should be no lower than the bottom of rap plate.

Emerge the edge of the homogenize bag opening at the door cover

levelly. After that, turn the door switch with you left hand until you

hear “Click”. By this time, the door and top rail seal the homogenize

bag by clamping it.

7. When the equipment finishes running, take out the homogenize

bag at its upper edge with your left hand and open the homogenizer

door with your right hand. Choose the rev again as you open the

homogenizer door.

8. You’d better stop all the panel operations or cut off the power for

your security.

ⅣⅣⅣⅣ Attentions:

1. Make sure the plug has been inserted into the socket tightly, any

looseness may result the computer control panel in a halt. If it is

halted, please switch off the rear panel and restart after 3min.

2. Do not attempt to open the homogenizer door when the rap plate is

running, otherwise, the sample liquid will splash. If you open the

door by force, the equipment would stop running automatically and

the display screen will stay still at that time. Then, close the door and

press the key , it will go on the remaining tasks automatically.

3. Please cut off the power and pull out the plug when you finish

using the equipment.

4. The bottom of the homogenizer door is empty to cope with

incidents, such as breakage of homogenize bag, and to wipe away

spilth easily, therefore, put your hands into the equipment from the

bottom while it is running is absolutely forbidden as your hands may

be injured.

5. Check whether the homogenize trunk is clean with no other

unnecessary matters before it goes to work, so that any accident and

damage to the homogenize bag can be avoided.

6. Any material of hard lump, bone or ice type are not suitable to be

put into homogenize bag as they can do damage to it.

7. Do not expose the equipment and homogenize bag to the sunshine

directly. To protect them from aging, please store in places with no

sunshine or no UV, especially homogenize bag.

6. Maintenance:

Clean the equipment, hammer plate, especially the homogenize bag

with clean cloth. Cleaning shall be carried out when power is switched

off.

7. Malfunction and Trouble Shooting

(1) If the machine fails to work after the door is soundly closed and

there is no display on the display screen, users shall examine the power

line to make sure it is connected normally and the fuse is in good

condition.

(2) If the machine sounds abnormal, examine bolts and hammer

plate to see whether it is soundly connected. If lubricant dries out, users

shall clean the hammer pole and hammer cover in time and lubricant

grease shall be re-filled in these parts.

(1) Under normal service and maintenance condition, end-users can

enjoy one year’s free maintenance and service from the day the machine

is delivered.

(2) When malfunction occurs, please deliver the machine to the HQ of

Ningbo Scientz Biotechnology Co., Ltd. Delivery payment shall be

prepaid.

As to functional disfigurement resulted from inappropriate use, or

operating and maintaining against the instrument or caused by

unauthorized staff, Ningbo Scientz Biotechnology Co., Ltd. will not take

on the responsibility to repair for free. Payment for repairing will be

charged according to specific conditions.

Examples (I)

Test Report of Granular Chinese Medicine

Date: October 13th, 2008

Equipment: scientz-09 sterilized homogenizer, rap plate coated with

rubber sleeve, silicon rubs of 2mm put inside the sides of glazed door.

The test includes two programs:

No. 1

1. Dip two of the granular medicine into boiled water for 10 min, and

then put them (medicine and water) together into a homogenize bag, after

that, put another homogenize bag outside the first one.

2. Finally, place the medicine of step 1 into the homogenizer to be raped

between the rubber sleeve and silicon rub for 2.5 min.

3. Rev: 6T/Sec

Result: No leakage occurs to both two homogenize bags.

No. 2

1. Dip two of the granular medicine into cold water for 10 min, and then

put them (medicine and water) together into a homogenize bag, after that,

put another homogenize bag outside the first one.

2. Finally, place the medicine of step 1 into the homogenizer to be raped

between the sleeve and silicon rub for 3 min.

3. Rev: 6T/Sec

Result: The inside bag is broken, outside one is normal, with uneven

surface, color of Program 1 is darker than Program 2.

Conclusion: Homogeneous effect can be achieved by dipping into cold

water for 10 min, with two homogenize bags and coating a silicon rub of

2mm.

Examples (II)

Obtain Large Scale of Individual Cells by Sterilized Homogenizer

Date: May 27th, 2008

Equipment: Scientz-09 Sterilized Homogenizer

Place the following tissues of tumor procured from operations on the

plate, such as gastric cancer, colon cancer and breast cancer, etc. Wash

them by physiological saline with 100U/ml gentamicin for three times to

remove bacteria on the tissues, and then add 10ml physiological saline

per 2g. Tear them into small pieces, as 2-5mm, afterward, put these pieces

into homogenize bags.

Operate as the instructions in the manual of sterilized homogenizer, as

8T/Sec for 2-8min, rest 1min after 1min’s operation, and record the

accumulative time. Different tissues may need different period of time.

Intake these tissue mixtures into test tubes for a standing of 3-5min, after

that, select out the clear cell suspension which includes a variety of

scattering individual cells, separate it for 5min with a centrifuge at the

speed of 1000 r/min. Add in 20% calf serum nutrient solution for culture

(directly is allowable) and control the cell population around 2×105ml.

Cells for this purpose are enough although the total population may have

change with the size of tissue.

These individual round cells in the following picture is the scattered

individual cells we needed, including glandular cells and epithelioid cells,

these irregular spindle cells are scattered myofibroblast, also known as

scattered individual cells. Both kinds of cells are intact and plump, with

excellent activity.
