Automation

Successful DNA typing depends on the quality and nature of the sample and purified DNA. The ideal DNA purification protocol separates DNA from the solid support, other biological material and potential PCR inhibitors and is amenable to automation for increased throughput.

Specialized allele-calling software eliminates the need for visual comparisons to assign alleles, making the DNA-typing process faster, easier and more accu-rate. However, analysis of STR data is more than allele calling. Expert systems software increases the efficiency or effectiveness of genotyping by automatically interpreting and reviewing STR data. Expert systems also can objectively evalu-ate DNA mixtures in seconds, flag potential contamination, sum marize data and generate reports.

DNA purification methods cannot differentiate between human DNA and contaminating bacterial and fungal DNA. Therefore, if the sample is not pristine and you want to de-termine the human genomic DNA concentration, you will need a human-specific DNA quantitation system. The most common systems are based on real-time PCR.

Drug DiscoveryIn a recent report, researchers at the NCGC describe use of P450-Glo™ Assays to monitor activity of small molecule library compounds stored at room-tempera-ture in DMSO or DMSO:water. They assayed CYP1A2 & 2D6 isoforms in a quantitative HTS format. The homo-geneous P450-Glo assay was faster and more efficient t han the traditional LC-MS method, and gave similar results.
