Each test kit contains a package insert with detailed information regarding the standard procedure of the measurement. In some cases, however, special points have to be considered which are not mentioned in the package inserts. In this appendix you will find specific applications and modified methods for analyses for sensitive measurements indicated with a 1).

Calcium: with Spectroquant® Calcium Test, Cat. No. 1.14815.0001 Measuring range: 1.0 – 15.0 mg/l Ca 2.5 - 37.5 mg/l CaCO3 1.4 - 21.0 mg/l CaO Procedure: Add 10 ml of reagent Ca-1 and 8 drops each of reagent Ca-2 and Ca-3 to 1 ml of the sample solution and mix. Leave to stand for 8 min for colour development. After filling into 10 mm cells measure in the photometer at 550 nm. Prepare the blank required for the measurement according to the above-mentioned procedure using 1 ml of distilled water instead of the sample solution. Procedure: Pipette 10 ml of sample solution into the reaction cell, mix and add 1 blue microspoon of reagent F-1K. Close the cell tightly and dissolve the solid by shaking. Leave to stand for 5 min for colour development. After filling into 50 mm cells measure in the photometer at 620 nm. Procedure: Place 1 microspoon of reagent SO3-1K into the cell, close tightly and shake until the complete dissolution of the solid. Then pipette 7 ml of the sample solution into the cell and mix. After the reaction time of 2 min fill into 50 cells and measure in the photometer at 410 nm. Prepare the blank required for the measurement according to the above-mentioned procedure using 7 ml of distilled water instead of the sample solution. Alternative methods 2)

Calibration curves for the methods are non-linear. 1. Use the below mentioned concentration-absorption-table 2. Beside using the concentration-absorption-table you can enter a non-linear curve by using the coefficients of

the following formula. C = a + bx + cx2 + dx3 Check your instrument and enter the coefficients according the given list. The calibration is always done against distilled water.
