The first enhancement is an expansion of the first submier by state report. This report represents the first time a pest/pathogen has been reported in a state to the NPDN National repository. Currently the system emails six different reports depending upon role access to regional and national program leader staff daily based on the previous day’s activities. An online report feature is also available for past dates. The enhancement will provide a similar report to the SPRO’s and diagnosticians with their specific state access. The enhancement will show all diagnoses from your state and all diagnoses your laboratory processed regardless of state of origin. The first release of this enhancement, with online report capability, will be at the end of 2010 and the email feature will be implemented in the first quarter of 2011. The second enhancement is the pest grouping feature. This item will support grouping by species, entries without common names, and a beer means to address synonyms. The pest/pathogen dictionary will still be available in its classic form and no software changes will be required for data entry on the part of the lab management systems. Most of the software for this effort has been developed, but assistance from a subgroup of the Database Program Area Commiee is required to accurately determine the list of unique scientific names and to do beta testing prior to its

AttheNPDNIT/Diagnosticians’jointworkgroupmeetinginOctober

threenewproposedenhancementswerepresented.Thefirstenhancement

Thirteen New State Detections of Duponchelia fovealisOnMay18,2010,theCanadianFoodInspectionAgency(CFIA)notifiedAPHISaboutaninterceptionofDuponcheliafovealislarvaeinanurseryshipmentfromSanDiegoCounty,California.OnJuly16,2010,APHISconfirmedadetectionatagreenhouseinVista,California.Trace-forwardinformationprovidedbytheaffectednurseryresultedintheexpansionofdetectionsurveysin26countieswithinCalifornia.

DuringthemonthsofSeptemberandOctober,APHISandStateDepartmentsofAgriculturehaveconfirmedthepresenceofDuponcheliafovealisinportionsoftwelveadditionalstatesincluding;Alabama,Arizona,Colorado,

Florida,Georgia,Mississippi,NorthCarolina,Oklahoma,Oregon,SouthCarolina,Texas,andWashington.

Atypeofmoth,DuponcheliafovealisoriginatedinsouthernEuropeandNorthAfricaandhasawiderangeofhostplants,includingpeppers,pomegranates,aquaticplants,beddingplantsandnurserystock.ReadthefullSPROletterhere.

RecentscientificliteratureidentifiesPseudomonassyringaepv.actinidiaeasadestructiveplantpathogenthatisseriouslyaffectingorchardsofkiwiplantsaroundtheworld.ThediseaseisrapidlyspreadingthoughAsiaandEurope.InItaly,itisestimatedthattheeconomiclosses(includingimpactontrade)duetoP.syringaepv.actinidiaehavereached2millionEuros.

Pseudomonassyringaepv.actinidiaeisnotknowntooccurintheUnitedStates.Plantpathogens,includingvirusesandviroids,areextremelydifficulttodetectduringaportofentryinspectionofthehostplants,particularlyintheabsenceofsymptoms.Thepathogenisnotknowntobeseedorfruittransmitted.

Diagnostic Updates

Loop-mediated Isothermal Amplification (LAMP) as a Diagnostic ToolMelodiePutnam,OregonState

AsadiagnosticianIamalwayslookingforassaysthatarerapid,

accurate,easytouse,andinexpensive.OnceIfoundoutitispossibletohavethespecificityofPCRwithouttheneedforthermalcyclers,gels,orexpensiveandshort-livedfluorescentprobesusingthemethodknownasLAMP,Iwasdefinitelyinterested.Loop-mediatedisothermalamplification(LAMP)takesplaceinasinglemicrofugetubeandrequiresonlyawaterbathorheatblock.Resultsareevaluatedvisuallywithoutextensivemanipulationofproduct.Sincethemethodistheequivalentofamusclecarasfarasspeed(fromafewcopiesofDNAto109,optimally,inlessthananhour)andishighlyspecific,itseemstohavethequalitiesthatwouldbeextremelyusefulinadiagnosticlaboratory.WehavedevelopedanassayfordetectionofRhodococcusfasciansusingthismethod(Serdani,etal.,2009),arecurrentlyusingLAMPforfireblightdetection(Templeetal.,2008),andhavebeenveryhappywithbothtests.Theassaycanbeusedonhosttissueinadditiontopurecultures,anditlivesuptotheclaimsforspeedandselectivity.Thefollowinginformationisprovidedtopiqueyourinterestindevelopingyourownassays.

LAMPwasdevelopedbyTsugunoriNotomiofEikenChemicalCompanyandcolleaguesattheUniversityofTokyoandOsakaUniversityMedicalSchool.Theirseminalpaperwaspublishedovertenyearsago(Notomietal,2000),andthemethodhassincebeenadapted

forusewithfungi(Tomlinson,etal,2010),nematodes(Kikuchi,etal.,2009),andviruses(Fukutaetal.2003)inadditiontophytobacteria.TheprocesstakesplaceinamicrofugetubecontainingasmallamountofextractedsampleDNAandfourprimersthatarespecifictosixregionsofyourtargetDNA.Astrand-displacingpolymeraseisusedat60-65°C(thespecifictemperatureuseddependsonsystemoptimization)andaseriesofloopedstructuresresult.TherathercomplexprocessofamplificationandthestructureformationisdetailedattheEikenwebsite,whichalsoincludesananimationofthereaction(http://loopamp.eiken.co.jp/e/lamp/index.html).Asaby-productofamplification,magnesiumpyrophosphateaccumulatesinproportiontotheamountofthespecificproductformed,andcanbeseenasturbidityinthereactiontube.Ifitisdifficulttoseethisturbidity,aDNAbindingdyemaybeused,whichwillintegratewiththeamplifiedproduct.Thisdye-DNAcomplexwillfluoresceunderUVlight,whichmakesiteasiertodistinguishbetweenpositiveandnegativereactions.

ForourLAMPassayforRhodococcusfascians,weuseeithertheFastSpinorQuiagenkitstoextractDNAwhenstartingwithplantmaterial,orifusingapureculture,simplyboilthebacteriabrieflytoextractDNA.Analiquotisthenaddedtothereactionmixture,thetubesareincubatedat60°C,andvisualizationiswithPicoGreen(orthefluorescentdyeofyourchoice).Theentireprocessis
