The DCE has r e s p o n s i b i l i t y f o r support o f b a s i c research and development e f f o r t s i n chemoprevention of cancer . As a program mechanism i n add i t ion t o ind iv idua l g r a n t sand con t r ac t s , t h e new P r o j e c t s are envisioned as means t o enhance and expandmultidiscipl inary/ interdisciplhary bas i c s t u d i e s i n development of new chemopreventive e n t i t i e s and s t r a t e g i e s f o r cancer prevent ion. Each NCCP would c o n s i s t o f a number of l abora to ry research programs represent ing d ive r se scientific d i s c i p l i n e s and e x p e r t i s e . S c i e n t i s t s in a given Pro jec t could de r ive from anycombination of t h e academic, non-profi t , and fo r -p ro f i t communities. S c i e n t i s t s i n an NCCP could a l s o be drawn from a s i n g l e organiza t ion possessing necessaryd i v e r s i t y and in-depth e x p e r t i s e t o accomplish P ro jec t ob jec t ives . Each P ro jec t is envisioned t o c o n s i s t of a Pro jec t Di rec tor , Program Leaders i n seve ra l broad s c i e n t i f i c d i s c i p l i n e s and an N C I Coordinator. Many c l a s s e s of chemopreventive agents have been inves t iga t ed in numerous b i o l o g i c a lsystems, and of t hese , a s i g n i f i c a n t number appear promising f o r s u b s t a n t i a l developmental e f f o r t s . These classes include, bu t are not l i m i t e d t o , p ro t easei n h i b i t o r s , an t iox idants , micronut r ien ts , calcium compounds, vitamin D, its metabol i tes and analogs, free r a d i c a l scavengers and i n h i b i t o r s of free r a d i c a l producing sequences, f a c t o r s nodulat ing growth and/or maturation, inc ludinglymphokines, modulators of arachidonic a c i d metabolism, nucleophi les and p o t e n t i a l new c l a s s e s of i n h i b i t o r s / suppressors e x i s t i n g i n n a t u r a l products such as foods consumed by man, as exemplified by green and yellow vegetables . In-depth,coordinated s t u d i e s are a l s o needed on t h e inf luence of macroconst i tuents of t h e d i e t on carcinogenesis . Since t h e r e is a l ready ex tens ive a c t i v i t y i n r e t i n o i d s research and development, app l i ca t ions i n t h i s area w i l l be considered non-responsive.

MECHANISM OF SUPPORT

Awards w i l l be made as Cooperative Agreements. These are a s s i s t a n c e r e l a t i o n s h i p s ,

involving s u b s t a n t i a l involvement by NCI s ta f f during performance of t h e P r o j e c t . The n a t u r e o f N C I staff p a r t i c i p a t i o n is included i n t h e RFA. However, t h e applying P r o j e c t must de f ine i t s o b j e c t i v e s i n accord with i ts own i n t e r e s t s and pe rcep t ions of novel approaches t o cancer prevention. The r o l e o f N C I s t a f f w i l l be t o provide a s s i s t a n c e , advice and guidance a f te r an award is made. F ina l decision-makinga u t h o r i t y dur ing performance w i l l rest with t h e P ro jec t D i rec to r .

N C I a n t i c i p a t e s t h e funding o f mul t ip l e awards f o r p r o j e c t per iods of f i v e ( 5 ) yea r s and has set a s i d e $1,500,000 f o r t h e i n i t i a l y e a r ' s funding. The expected s t a r t i n g d a t e f o r t h e s e awards is September 1, 1987. Although t h i s program is provided f o r *

BACKGROUND tThe Divis ion o f Research Resources ( D R R ) is continuing i ts competit ive Biomedical Research Support (BRS) Shared Ins t rumenta t ion Grant Program i n i t i a t e d i n F i s c a l Year 1982. The program was e s t a b l i s h e d i n recogni t ion o f t h e long-standing need i n t h e biomedical r e sea rch community t o cope with r ap id technologica l advances i n ins t rumenta t ion and t h e r ap id ra te of obsolescence o f e x i s t i n g equipment. The o b j e c t i v e of t h e program is t o make a v a i l a b l e , t o i n s t i t u t i o n s with a highconcent ra t ion of PHS-supported biomedical i n v e s t i g a t o r s , research instruments which can only be j u s t i f i e d on a shared-use b a s i s and f o r which meritopious r e sea rch p r o j e c t s are desc r ibed ,

An e l i g i b l e i n s t i t u t i o n may submit more than one app l i ca t ion f o r d i f f e r e n tins t rumenta t ion f o r t h e February 17, 1987 deadl ine . However, i f mu l t ip l ea p p l i c a t i o n s are submitted f o r similar ins t rumenta t ion from one o r more e l i g i b l ecomponents o f an i n s t i t u t i o n , t hen documentation from a high admin i s t r a t ive o f f i c i a l must be provided, s t a t i n g t h a t t h e mul t ip l e a p p l i c a t i o n s a re 'a coordinated i n s t i t u t i o n a l resource p lan , no t an unintended dup l i ca t ion .

RESEARCH GOALS AND SCOPE

This program is designed t o meet t h e s p e c i a l problem o f a c q u i s i t i o n and updating of expensive shared-use instruments which are not gene ra l lv a v a i l a b l e through o t h e r PHS mechanisms, such as t h e r egu la r research , program p r o j e c t and c e n t e r g ran t programs, o r t h e Biomedical Research Support (BRS) Grant Program. Proposals f o r t h e

si development of new ins t rumenta t ion w i l l no t be considered.

ELIGIBILITY

The BRS Shared Ins t rumenta t ion Grant Program is a subprogram of t h e BRS Grant Program o f DRR. Awards are made under t h e a u t h o r i t y of t h e BRS program and are made t o i n s t i t u t i o n s only, no t t o ind iv idua l s . Therefore, e l i g i b i l i t y is l i m i t e d t o i n s t i t u t i o n s which r ece ive a BRS g ran t award. Awards are contingent on t h e a v a i l a b i l i t y of funds.

MECHANISM OF SUPPORT

BRS Shared Instrumentat ion Grants provide support f o r expensive s ta te -of - the-ar t instruments u t i l i z e d i n both bas i c and c l i n i c a l research . Appl ica t ions are l i m i t e d

/ t o instruments t h a t c o s t a t least $100,000 per instrument o r system. Types of ins t rumenta t ion supported include, bu t are not l i m i t e d t o , nuc lear magnetic

i-urv resonance systems, e l e c t r o n microscopes, mass spectrometers , p r o t e i n sequencer/aminoa c i d ana lyzers and ce l l s o r t e r s . Support w i l l not be provided f o r genera l purposebquipment or pure ly i n s t r u c t i o n a l equipment. Proposals f o r "stand alone" computer systems w i l l on ly be considered i f t h e instrument is s o l e l y dedica ted t o t h e research needs o f a broad community of PHS-supported i n v e s t i g a t o r s .

Awards w i l l be made f o r t h e d i r e c t c o s t s of a c q u i s i t i o n o f new, or t h e updat ing o f e x i s t i n g , research instruments . The i n s t i t u t i o n must meet those c o s t s (no t covered j.n t h e normal purchase p r i c e ) requi red t o p l ace t h e ins t rumenta t ion i n ope ra t iona lo rde r as w e l l as t h e maintenance, support personnel and s e r v i c e c o s t s a s soc ia t ed wi th maximum u t i l i z a t i o n o f t h e instrument . There is no upper l i m i t on t h e c o s t o f t h e instrument , bu t t h e maximum award is $300,000. Grants w i l l be awarded f o r a per iod of one year and are not renewable. Supplemental a p p l i c a t i o n s w i l l no t be accepted. The program does not provide i n d i r e c t c o s t s or support f o r cons t ruc t ion or a l t e r a t i o n s and renovat ions. Cost shar ing i s not requi red . If t h e amount of funds requested does not cover t h e t o t a l c o s t o f t h e instrument, an award w i l l no t be made un le s s t h e remainder of t h e funding is assured. Descr ip t ion o f t h e proposedco-funding must be presented with t h e app l i ca t ion . Assurance o f co-funding, s ignedby an appropr i a t e i n s t i t u t i o n a l o f f i c i a l , must be presented t o DRR p r i o r t o t h e issuance o f an award.

A major u s e r group o f t h r e e or more i n v e s t i g a t o r s should be i d e n t i f i e d . Each major u s e r must have PHS peer-reviewed research support a t t h e t i m e of t h e award. The a p p l i c a t i o n must show a clear need f o r t h e instrumentat ion by p r o j e c t s supported bymul t ip l e PHS research awards and demonstrate t h a t t h e s e p r o j e c t s w i l l r e q u i r e a t least 75% o f t h e t o t a l usage of t h e instrument . Major u s e r s can be ind iv idua l r e sea rche r s , or a group of i n v e s t i g a t o r s wi th in t h e same department o r from s e v e r a l departments a t t h e app l i can t i n s t i t u t i o n . PHS extramural awardees from o the r i n s t i t u t i o n s may a l s o be included.

If t h e major u s e r group does not r e q u i r e t o t a l usage of t h e instrument, access t o t h e instrument can be made a v a i l a b l e t o o the r u s e r s upon t h e advice o f t h e advisorycommittee. These users need not be PHS awardees but p r i o r i t y should be given t o PHS supported s c i e n t i s t s engaged i n biomedical research .

The Divis ion o f Epidemiology and C l i n i c a l Applicat ions (DECA) of t h e Nat ional Heart,Lung, and Blood I n s t i t u t e (NHLBI) has i n i t i a t e d t h e Prevent ive Cardiology Academic Award (PCAA) t o provide a s t imulus f o r t h e development o f a prevent ive card io logycurriculum i n those schools of medicine and osteopathy t h a t do not have one and t o s t rengthen and improve t h e prevent ive cardiology curriculum i n t h s s e schools t h a t do. Each school of medicine or osteopathy i n t h e United S t a t e s and its possess ions or t e r r i t o r i e s is e l i g i b l e t o compete for one award f o r a p r o j e c t per iod t h a t does not exceed f i v e yea r s . The number of awards made each year w i l l depend upon t h e m e r i t of t h e a p p l i c a t i o n s received and a v a i l a b i l i t y of funds.

For t h e purposes o f t h e PCAA, t h e term prevent ive cardiology is used t o d e f i n e t h e area o f card iovascular medicine having a s p e c i a l concern with t h e development o f knowledge and t h e a p p l i c a t i o n of knowledge d i r e c t e d a t t h e prevent ion o f h e a r t and vascular d i seases . The National I n s t i t u t e o f Dental Research ( N I D R ) i n v i t e s app l i ca t ions f o r t h e support of c a t e g o r i c a l Per iodonta l Diseases Research Centers t o conduct m u l t i d i s c i p l i n a r y , fundamental and c l i n i c a l research on t h e e t io logy , d iagnos is ,pathogenesis, t rea tment and prevention o f per iodonta l d i seases .

BACKGROUND

The NIDR is c u r r e n t l y suppor t ing f i v e per iodonta l research c e n t e r s whose approvedfunding pe r iods w i l l end i n t h e summer o f 1988. The N I D R expec ts t o continue t h i s program o f pe r iodon ta l research c e n t e r s a t l eas t 10 yea r s beyond 1988. The p resen t announcement e s t a b l i s h e s a new round o f competit ion f o r t h e f i v e year per iod of funding from 1988 t o 1993. This competit ion w i l l be open t o a l l domestic i n s t i t u t i o n s , inc luding those proposing new per iodonta l research cen te r s , as w e l l as t h o s e with e x i s t i n g c e n t e r s . It is a n t i c i p a t e d t h a t a new RFA w i l l be i ssued subsequently f o r t h e per iod from 1993-1998.

RESEARCH GOALS AND SCOPE

One o f t h e goa l s of t h e N I D R is t o accelerate and expand t h e development o f new information which may l e a d t o t h e prevention and c o n t r o l o f per iodonta l d i s e a s e s . To achieve t h i s goal and t o provide g r e a t e r focus f o r per iodonta l research , it is e s s e n t i a l t h a t t h e N I D R maintain a program o f s t rong , mu l t id i sc ip l ina ry c e n t e r s capable of pursuing promising l e a d s i n an in t eg ra t ed fash ion , us ing modern molecular biology and o t h e r new technologies , as w e l l as more t r a d i t i o n a l approaches. To provide adequate a t t e n t i o n t o all research areas be l ieved t o have s i g n i f i c a n t p o t e n t i a l , t h e NIDR seeks t o develop a balanced o v e r a l l program i n which t h e pro toco l s o f t h e d i f f e r e n t c e n t e r s are complementary r a t h e r than dup l i ca t ive . Col labora t ive arrangements between c e n t e r s w i l l be encouraged. I n June, 1985, t h e Div is ion o f Epidemiology and C l i n i c a l Appl ica t ions(DECA) he ld a Workshop on 24-Hour Ambulatory Blood Presnure Monitoring (ABPM), which i d e n t i f i e d a number o f r e sea rch needs t o f i l l gaps i n our knowledge concerningappropr i a t e u ses o f t h i s promising technology and t o r e f i n e t h e ins t rumenta t ion and methods f o r d a t a a n a l y s i s .

The purpose o f t h i s Program Announcement i s t o encourage i n v e s t i g a t o r s t o submit r e sea rch g r a n t a p p l i c a t i o n s involving t h e use o f 24-Hour ABPM. Some o f t h e areas where a d d i t i o n a l r e sea rch is needed are l i s t e d below:

1 Charac te r i za t ion o f t h e normal 24-hour ABPM p r o f i l e us ing c u r r e n t l ypublished methods and ana lyses i n r e p r e s e n t a t i v e samples o f normotensive ind iv idua l s , inc luding women and minor i t i e s ; a l s o de te rmina t ion o f t h e optimalfrequency and du ra t ion of ABPM f o r s p e c i f i e d purposes.
