Generate high resolution molecular karyotypes in a single experiment with PerkinElmer’s array CGH technology and software from Spectral Genomics.Chromosomal abnormalities (deletions, duplications, and amplifications) are traditionally determined using whole chromosome preparations from cells (metaphase spreads). These are analyzed using G banding, FISH or by chromosomal Comparative Genomic Hybridization (CGH).
These traditional methods for detecting chromosomal abnormalities are well established, but they do not provide the speed, throughput, or resolution required for today’s cytogenetic laboratories. They are also time consuming and cumbersome, and require the skills of a trained cytogeneticist to analyze and interpret.
Rapid Analysis with High ResolutionPerkinElmer’s Spectral Genomics array CGH technology(aCGH) circumvents the considerable limitations of usingmetaphase spreads and takes karyotyping to a new level of performance and ease-of-use.
Traditional array-based methods for detecting genomic variation, such as SNP and oligo arrays often yield resultsthat are noisy, or unclear. Our Spectral GenomicsSpectralChip™ 2600 BAC arrays provide researchers with high quality results that facilitate genetic disease research.Because each BAC is printed in duplicate, and a dye swapreaction is performed, you obtain reliable results.
The SpectralChip 2600 allows investigators to rapidly pan for chromosomal biomarkers across the entire humangenome. You can generate a quantitative genome-widemolecular profile of chromosomal imbalances on a singlechip. Research applications* include:

Invented by PerkinElmer scientists, TSAcan be used in any application allowingthe addition of horseradish peroxidase(HRP) into its protocol, such as immuno-histochemistry, in situ hybridization,ELISA and microarray-based differentialgene and protein expression studies.
In standard IHC and ISH protocols, TSAamplifies both chromogenic and fluores-cent signals, resulting in a significantincrease in sensitivity, without loss of resolution or increase in background. Use it in immunohistochemistry or ELISAapplications, and unamplified detectionlevels can be maintained while using upto 1,000-fold less primary antibody.
Innovative Amplification ApproachMaintains Signal Clarity
TSA is easily integrated into any protocolafter an initial addition of horseradishperoxidase (HRP). The HRP is used to catalyze the deposition and binding of alabeled (e.g., biotin, DNP or other labelingmoieties) tyramide onto tissue sections orcell preparations previously blocked withproteins. This reaction is quick (less than10 minutes). The binding is covalentbecause the reaction intermediatelydimerizes with tyrosine residues on thesurface-bound endogenous proteins. Theselabels can then be detected by standardchromogenic or fluorescent techniques.Since the added labels are only depositedproximal to the enzyme site, there is minimal, if any, loss of resolution.

PolyScreen PVDF Transfer MembranesA microporous polyvinylidene difluoride membrane, PolyScreen is an effective alternative to nitrocellulose membranes for protein transfers.
Sensitive: detects low levels of protein and decreases exposure times.
Easy to use: adapts to most nitrocellulose membrane protocols with the addition of a pre-wetting step.
Durable: stronger than nitrocellulose, it won’t tear, crack or shrink, so there are no rips or crackles on your autorads and your molecular weight markers are not distorted.
Reprobable: retains more protein than nitrocellulose, even during the most stringent washes.

Tyramide Signal Amplification (TSA) SystemsPerkinElmer’s patented Tyramide Signal Amplification(TSA) technology, extensively referenced in scientific literature, is widely recognized as a valuable tool for amplifying both chromogenic and fluorescent signalsobtained with standard immunohistochemistry (IHC) and in situ hybridization (ISH) protocols.
Our TSA systems are formatted to enable every lab to benefit from the sensitivity enhancements made possible by applying this innovative technology.
Benefits in Use• Provides a significant increase in sensitivity, without
loss of resolution or increase in background.
• Eliminates need for IS-PCR (target amplification) sometimes used prior to ISH assays when low or single copy genes are present.
• Utilizes up to 1,000-fold less primary antibody and main-tains detection levels, when used in IHC assays, ofteneliminating the need for antigen retrieval procedures.
• Conserves limited or expensive probes when added to ISH protocols.
• Enables visualization of previously undetectable levels of protein or nucleic acid, possibly leading to newresearch insights or discoveries.
Choosing a TSA SystemChoose a system that best suits your research needs:
• Select chromogenic or fluorescence detection based on the equipment available to your laboratory and your need for a permanent record.
• Eliminate biotin interference: consider the level ofendogenous biotin in your specimens and your desire to eliminate biotin-quenching procedures.
• Choose from several fluorophores based on your preferredfluors and your interest in performing multi-target detection.
• Determine the degree of amplification you need, takinginto account the abundance of the protein or gene in yourspecimen and your need to conserve antibody or probe.
TSA is compatible with all common chromogenic and fluo-rescence detection methods, and is easily integrated into anyprotocol after an initial addition of horseradish peroxidase(HRP). Based on your experience and sensitivity require-ments, one of three formats will best fit your needs:
Complete Systems
• TSA Systems: complete tyramide signal amplificationkits for IHC or ISH applications.
• TSA Plus Systems: tyramide signal amplification kits thatprovide 10 to 20 times the sensitivity enhancement of regular TSA systems. Available in a biotin-free DNP alternative for chromogenic detection.
Reagent Packs
• TSA Reagents: stand-alone amplification reagents thatprovide researchers experienced with TSA the flexibilityto develop their own protocols.
Complete TSA SystemsAll TSA systems include tyramide labeled with either a haptenor a fluorophore, amplification diluent and a comprehensiveinstruction manual. TSA Plus Fluorescence Systems provide very high sensitivityin ISH compared to traditional methods. While developedspecifically for ISH applications, these systems can also beused with IHC applications.

Spectral Genomics SpectralChip 2600 SpectralChip 2600 arrays are 2600-element BAC arrays used to evaluate CGH or chromosomal abnormalities at locations approximately 1 megabase apart across the entirehuman genome. Our human BAC arrays generate a genome-wide molecular profile and quantification of chromosomalimbalances on a single chip.
Our arrays display high sensitivity with low background,facilitating image analysis for the qualitative analysis ofhybridization. DNA (obtained from blood, amniotic fluid,chronic villi or tissue samples) is analyzed directly byhybridization.

SpectralWare software accepts data from a variety of arrayscanners, including PerkinElmer’s ScanArray product line. It converts two-color fluorescent dye signals into intensityratio profiles. The profiles for each chromosome are correlatedand displayed with ideograms of the chromosome to showthe loci of each feature detected in the genome. SpectralWaredisplays chromosomal deletions and amplifications in aunique graphic presentation for rapid interpretation of results.
