A mouse monoclonal antibody, clone HCD54, specific for human CD54 was purchased from a commercial source in both unconjugated and RPE or FITC conjugated formats. The unconjugated antibody was linked to RPE or FITC using a Lightning-Link® kit, and the two conjugates were compared in flow cytometry staining human peripheral blood lymphocytes, where the blue curve shows unstained cells. Lightning-Link® conjugated antibody is shown in the middle, and commercially conjugated antibody on the left hand side. The right hand histograms show an overlay of the positively stained cells.

Anti-human IgG monoclonal antibody was purchased pre-conjugated to alkaline phosphatase from a commercial supplier. The same antibody was purchased in un-conjugated form and then conjugated to alkaline phosphatase using a Lightning-Link® kit.

The Lightning-Link® conjugate demonstrates enhanced titre and sensitivity.

A comparison of antibody conjugation using InnovaCoat® GOLD against that using traditional passive conjugation techniques, with naked gold colloid, showing enhanced signal intensity with InnovaCoat® GOLD. Anti-IgA antibodies were labelled with 40nm gold particles, and used to measure IgA concentration in a competitive lateral flow inhibition assay: IgA in the sample competes with that bound to the nitrocellulose on the lateral flow strip and therefore as the concentration in the sample increases the signal intensity decreases.

The data in the graphs above demonstrates the enhanced signal intensity observed when using InnovaCoat® GOLD in comparison to passively conjugated naked gold, under identical conditions. The curve resulting from the use of InnovaCoat® GOLD provides significantly increased sensitivity for measuring the IgA concentration. Furthermore, the covalent attachment of the antibody to the InnovaCoat® GOLD provides enhanced stability and prevents aggregation of the conjugate.
