INTRODUCTIONHeterocyclic diamidines, and related compounds with an appropriate structure can bind strongly to the DNA minor groove and exhibit important biological effects including clinical antiparasiticactivity

Recent results show that thecompounds can compete withtranscription factor binding in boththe minor and major groove

The compounds can also target the AT-rich sequences in mitochondrial kinetoplatsminicrcle DNA

GENERAL OBJETIVE

Demostrate that thetopologicalchanges in DNA minicircles induced by compoundsthat bind to the minor groove, can lead to the destruction ofthe kinetoplast duringopening of the minicircles fortranscription and replicationand therefore, play a role inthe antiparasitic activity of thecompounds

RESULTADOS FIGURA 2

A5 muestran un aumento de la movilidad (fv)

Las secuencias ATATA tienen una movilidad

reducida (fa)

DB75 no efectos topológicos detectables

DISCUSSION

WHAT HE SAIDIS HE

Tevis D.S, et

al.

Experiments with ligation ladders from

DNA sequences with AAAAA and ATATA

binding sites have shown that DB75 and

some close analogs induce topological

changes in DNA that are sensitive to DNA

sequence/structure and to compound

structure and properties

AND

Paine,M.F,

et al.

At present there is considerable

knowledge of the effects of protein binding

on DNA topology but much less

information about small molecules that

target DNA. Such compounds are being

designed for therapeutic applications that

range from antiparasitic to anticancer

activities

DISCUSSION

WHAT HE SAIDIS HE

AGREE?

Soeiro,M.N. et

Topological changes on binding of the

heterocyclic diamidines of Figure 1 to

the mitochondrial kinetoplast DNA of

disease-causing eukaryotic parasites

may be a key step in destruction of the

kinetoplast and the biological

mechanism of action of the

compounds

YES

Munde,M, et

al.

This could be due to the fact that

compounds, such as DB1003 and

DB293, have been shown to bind to the

TTAA sequence as a dimer

CONCLUSIONS

This study is very important to evit some

antiparasitic human deseases since ADN

structure

We have to see deseases since the

molecular point of view, because i think

its easilier to trate
