A POWERFUL NEW GENERATION OFDIAGNOSTIC TOOLSThe HistoSondas® are new single stranded DNA probes for chromogenicvisualization of gene expression by in situ hybridization.

The characteristics of these innovative new probes set them apart fromthose already on the market.

The HistoSondas® consist of single stranded DNA fragments whoselengths are between 10 and 30 times greater than commercial oligo probes(the length of a HistoSonda® is between 200 and 1300 nucleotides).

The probe DNA is targeted against RNA expressed by genes of interestand therefore the HistoSondas® are probes for the detection of geneexpression. The HistoSondas® are specifically designed to be used onformalin-fixed and paraffin-embedded tissue sections.

is fasterThe HistoSondas® have an incubation temperature of 62ºC. The elevatedthermal energy generated allows the molecules to vibrate quicker thereforetaking less time to find their target even though they are situated in a veryviscose dextran containing media. The use of 62ºC as the incubationtemperature for these probes therefore permits rapidity. HistoSonda® is theonly commercial probe whose incubation period is just 30-60 minutes.

is more specificDue to their great lengths the HistoSondas® are more specific. It is statisticallyless probable to find shared sequence homology between long sequences.Therefore the chances of the probe annealing with anything other than thetarget sequence are greatly reduced, increasing specificity. The HistoSonda®incubation temperature of 62ºC also permits that only chains withalmost total sequence homology are annealed (an oligo will not anneal at thistemperature).

is more potentThe potency of the HistoSondas® is directly influenced by the number oflabeled uridines in the DNA sequence. As the HistoSondas® have longsequences, the proba-bility of large num-bers of labeleduridines being incor-porated is greatly in-creased, increasingthe potency of thesignal.

is simple to use

The HistoSonda® hybridization procedure has been simplified to themaximum. It only uses one buffer during the entire technique (PBS). The useof formamide is not necessary and post-incubation washes with formamidehave therefore been suppressed. The hybridization procedure can be carriedout without needing to have specific knowledge of Molecular Biology.Complicated apparatus is not necessary and revealing the probe can beperformed automatically using any immunohistochemistry apparatus.

The Histosondas® are supplied Lyophilized in single use tubes. Transport andstorage can be carried out at room temperature and they have a long storagelife. Freezers, refrigerators or any special type of care are not required.
