Laboratory Scale Tide Motion Bioreactor: CelCradleTM

CelCradleTM is a cost-effective, single-use,benchtopbioreactorsystemcapableofsupportingthehigh-densitycultureofadherentoranchorage-dependentcells. It isdesignedbasedon theconceptofbellow-inducedintermittentflowofmediaandairthroughporousmatriceswherecellsreside.Thisprovidesalowshearstress,highaeration,andfoam-freecultureenvironment.

During operation, the CelCradle™ bottleis partially filled with culture medium andinoculatedwithcells.Theculturemedium israisedand loweredalternately, to submergeinculturemediumandexposethematricestooxygen,creatingadynamicinterfacebetweenairandmediaonthecellsurfacetomaximizenutrient uptake andoxygen transfer.Whenmatricesexposetoair,cellsuptakeoxygenwhileprotectedbythinlayerofmediainmatrices.

AutoFeedersystemhasthreemajorcontrolfunctions:automaticcontroloffeedandharvestvolumebyweightcalculation,pHcontrol,anddissolvedoxygen(DO)levelcontrol.

Duringfeedingandharvesting,twolargeperistalticpumpsareusedtopumpmediuminandoutataspeedof2,000ml/min.Thisallowsupto900Lofculturemediumtobetransferredperday,whichcanbedividedinto1–96cycles.Astepprogramsystemcanbeusedtosetmediumexchangevolumeinastep-wisemanner.Userscansetthemediumexchangevolumeateachstepandcancreateuptotendifferentsteps.

ForpHcontrol,asmallpumpisusedforthealkaliaddition.Theusercansetthesamplingandflushingtime,aswellasthelowerandupperlimitsofthealarm.DOleveliscontrolledbytheAutofeederthroughlevelsofairandoxygen.

AutoSwitch System

TheAutoSwitchSystemisrecommendediftherearetwofeedingtanksusedduringperfusionculture.Itisconnectedinbetweenthetwofeedingtanks.Thepinchvalveinsidethesystemwilldetectbubbleandautomaticallyswitchfromonetanktoanotheroncethefirstreservoirisempty.Analarmlightwillturnononcethemediainthetankisdepleted;therearetwoindividualalarmlightstoindicatewhichfeedtankisempty.Also,theAutoSwitchSystemincludesin-lineandnon-invasivebubbledetector.

TideCell® Cell Harvesting System
