About
The JuLI™ FL, is the next generation of a smart fluorescence live cell movie
analyzer & counter.
The JuLI™ FL uses state-of-the-art optics to get live-cell images from
various cell culture dishes and can be performed cell-based assays, including
cell counting, cell viability and GFP/RFP expression level checking.
This also enables user to support the quantified cell confluence results with low
variation and the growth curve using image based analysis for bright field.

NIH-3T3 cells growth were observed 50 hours through JuLI™ Br. All images were recorded every 10 minute
and analyzed monolayer confluence.

Cell-growth images were captured for 63 hours with 20 minutes intervals in U-2 OS GFP & RFP stable cell line.
Live cell image capture
U-S OS cells were transfected using the Neon™ transfection system (from Life Technologies) and 0.25 µg of the
the EGFP-N1 plasmid & RFP plasmid. 48 hours after transfection, the images are captured.

U-2OS stable cells were incubated for 100 hours after scratch. JuLITM FL calculated the confluence with growth
of surface unfarmed automatically.

Cell counting & viability
Clumped cell (HepG2, MCF7, F9) cells were counted with a JuLITM FL, a hemocytometer and the competitor’s automated
cell counter. Accuracy and precision are comparable between the JuLITM FL and the hemocytometer for all cell lines.
Cell counting results were performed on three different instruments with seven sample replicates
Fluorescence expression level checking
JuLITM FL counting software provides actual cell counts, viability and automatically calculates cell concentration.
It also can identify and measure the levels of fluorescence (GFP or RFP expression).

GFP expressing cells (U-2 OS) were counted using JuLITM FL counting software (A). The histograms in
graph (B) show the green fluorescence intensity of the cells.

JuLI Br VS FL 기능적 개선 사항
•Capture and monitoring 시 다양한 image format: Br현재 (JPEG) à FL (JPEG, TIFF, BMP, PNG)로 확장 장점: 다양한 포멧 제공 가능, PC 베이스처럼 확장 함. 단점: 이미지 포멧에 따른 용량이 상이하게 차이 남 – data transfer 시 상당한 시간 소요됨
•모니터링 시 세팅 조건 변경: Br현재 (모니터링 한번 설정 하면 세팅 조건 변경 안됨) à 모니터링 시 일시 정지 후 Focus 탭으로 이동하여 Focus / Brightness / Exposure 재 조정 가능 장점: 몰폴로지 변화 등으로 초점이 나가거나 사용자가 세팅 조건 조정 가능 단점: Cell growth curve 그릴때 세팅 조건을 봐꾸면, 그래프가 연속되지 않고 출렁 거리거나, 끊어지는 것 처럼 보일 수 있음 (이는 사용자 조건 변화에 따른 것으로 complaint 아님)
