T h i s p r o j e c t w i l l c o n s i s t o f a double-bl ind randomized t r i a l and i s t o be c a r r i e d o u t by 2-4 c l i n i c a l c e n t e r s and a d a t a c o o r d i n a t i n g c e n t e r . An a t tempt w i l l be made t o r e c r u i t a t l e a s t 5,400 e l i g i b l e women so t h a t t h e r e w i l l be a f i n a l s t u d y sample of a t l e a s t 3,600. O f f e r o r s w i l l be expec ted t o i n c l u d e p l a n s f o r t h e r e c r u i t m e n t o f e l i g i b l e m i n o r i t y women f o r t h i s s t u d y . Biomedical r e s e a r c h w i l l be most e f f e c t i v e l y advanced by t h e cont inuous a p p l i c a t i o n o f a combinat ion o f models i n a complementary and i n t e r a c t i J e manner, r a t h e r t h a n by c o n c e n t r a t i n g on any one o r a few k i n d s o f model systems. Each system, whether mammalian o r nonmammalian, h a s i t s own uniqueadvantages and l i m i t a t i o n s . Because of a cont inued , wide i n t n r e s t i n t h e development o f nonmammalian models f o r biomedical r e s e a r c h , t h . p. work on systems t h a t a r e p h y l o g e n e t i c a l l y more c l o s e l y r e l a t e d t o humans. Experience i n d i c a t e s t h a t in format ion y i e l d e d by such systems can i n c r e a s e s u b s t a n t i a l l y t h e knowledge o f human f u n c t i o n .

Animals a r e e s s e n t i a l t o t h e advancement o f knowledge i n t h e biomedical s c i e n c e s . Nonmammalian r e s e a r c h methods can, and do, provide a d d i t i o n a l o p p o r t u n i t i e s t o advance our unders tanding o f b i o l o g i c a l p r o c e s s e s . For example, b i o l o g i c a l models o r model systems d e r i v e d from, o r c o n s i s t i n g o f ,nonmammalian organisms, o r c e l l and t i s s u e c u l t u r e systems, may providev a l u a b l e i n s i g h t s i n t o mechanisms o f b i o l o g i c a l f u n c t i o n s t h a t are more d i f f i c u l t t o o b t a i n from s t u d i e s o f whole v e r t e b r a t e an imals . Such t e c h n o l o g i e s o f t e n permi t s t u d i e s t h a t o t h e r w i s e would be imposs ib le t o perform. Mathematical modeling i s a n o t h e r u s e f u l i n v e s t i g a t i o n a l s t r a t e g ywhen c l o s e l y coupled t o b i o l o g i c a l exper imenta t ion , and t h e r e are o p p o r t u n i t i e s f o r mathematical modeling i n many a r e a s o f biomedical r e s e a r c h . Non-invasive exper imenta l t e c h n i q u e s , such as t h o s e commonly used f o r human s u b j e c t s , which permi t s t u d i e s of b i o l o g i c a l p r o c e s s e s i n i n t a c t an imals , can reduce t h e number o f exper imenta l an imals s i n c e m u l t i - s t e p phenomena can be observed i n a s i n g l e s u b j e c t .

Many s t r a t e g i e s a r e c u r r e n t l y i n p l a c e t o reduce t h e p a i n and d i s t r e s s of l a b o r a t o r y an imals ; however, new methods and t e c h n o l o g i e s are encouraged.

RESEARCH GOALS

Grant a p p l i c a t i o n s are r e q u e s t e d f o r p r o j e c t s t h a t w i l l i n c r e a s e t h e e x t e n t and depth o f knowledge needed t o develop methods o f biomedical r e s e a r c h t h a t :

o do n o t r e q u i r e t h e u s e of v e r t e b r a t e an imals o reduce t h e number o f v e r t e b r a t e an imals used i n r e s e a r c h o produce l e s s p a i n and d i s t r e s s i n v e r t e b r a t e an imals t h a n methods c u r r e n t l y used o v a l i d a t e o r demonstrate t h e r e l i a b i l i t y of non-animal methods o expand n o n - v e r t e b r a t e animal r e s e a r c h methods t h a t have been found v a l i d and r e l i a b l e

MECHANISM OF SUPPORT

The suppor t mechanism f o r t h i s program w i l l be t h e t r a d i t i o n a l ,. i n v e s t i g a t o r - i n i t i a t e d r e s e a r c h p r o j e c t g r a n t . Under t h i s mechanism, t h e

a p p l i c a n t w i l l p l a n , d i r e c t , and c a r r y o u t t h e r e s e a r c h program. The p r o j e c tp e r i o d d u r i n g which t h e r e s e a r c h w i l l be conducted should a d e q u a t e l y r e f l ec t t h e t i m e r e q u i r e d t o accomplish t h e s t a t e d g o a l s and be c o n s i s t e n t w i t h t h e p o l i c y f o r g r a n t s u p p o r t . Support w i l l be provided f o r up t o f i v e y e a r s(renewable f o r subsequent p e r i o d s ) s u b j e c t t o t h e a v a i l a b i l i t y o f funds and p r o g r e s s achieved .

Research g r a n t a p p l i c a t i o n s may b e submi t ted by both n o n p r o f i t and prof i t -making o r g a n i z a t i o n s and i n s t i t u t i o n s , S t a t e o r l o c a l governments and t h e i r a g e n c i e s , and e l i g i b l e a g e n c i e s of t h e Federa l Government.

APPLICATIONS AND REVIEW PROCEDURES
