CD133 is a cell-surface protein that identifies neural stem cells, which give rise to neurons and glial cells. This antibody is expressed on primitive hematopoietic stem and progenitor cells, retinoblastoma, hemangioblasts, and neural stem cells as well as on developing epithelia.

CD117 is expressed on the surface of hematopoietic stem cells as well as other cell types. This receptor binds to stem cell factors.

This 90 kDa disulfide-linked homodimer is expressed by vascular endothelial cells. It is suggested that CD105 functions in adhesion and embryonic angiogenesis.

Functionally, stem cells are able to propagate and generate additional stem cells, and can differentiate into various progenitor cells which further maturate along specific lineages. Stem cell markers allow for the identification and isolation of stem cells in different types of tissues and cell populations. Such markers allow identification of embryonic, hematopoietic, mesenchymal/stromal, and neural stem cells. Embryonic stem (ES) cells, derived from the inner cell mass of preimplantation embryos, are recognized as

the most pluripotent population, having the greatest developmental potential. However, for many applications adult stem cells may be preferred due to ethical issues involved in the derivation of ES cells. Stem cell research is poised to allow the development of new therapies through repair, replacement, and regeneration of damaged or diseased cells and tissues making it one of the most exciting frontiers in modern medicine.

The cripto gene is expressed both in embryonic stem cells and during the early phases of embryo development, while, in the adult, it is reactivated in a wide range of epithelial cancers.

The highly glycosylated 75-120 kD antigen CD34 is possibly an adhesion molecule with a putative role in early hematopoiesis by mediating the attachment of stem cells to the bone marrow extracellular matrix or directly to stromal cells. It could act as a scaffold for the attachment of lineage specific glycans, allowing stem cells to bind to lectins expressed by stromal cells or other marrow components. Staining for CD34 has been used to measure angiogenesis, which reportedly predicts tumor recurrence.

Connexin 43 is a negative cell surface marker for the stem cell-containing population of human limbal epithelial cells. Connexin 43 forms gap-junctional channels and regulates the permeability of these gap junctions to small organic molecules. Gap junctional intercellular communication is thought to play a key role in development and may also be involved in epilepsy.

Nestin is a Class VI intermediate filament expressed in the developing CNS in early embryonic neuroepithelial stem cells. This protein has been widely used as a predominant marker for stem/progenitor cells, glioma cells, and tumor endothelial cells.

The p63 gene is highly expressed in the basal or progenitor layers of many epithelial tissues. p63 plays a fundamental role associated with development and differentiation.

Podocalyxin is a CD34 family member expressed by podocytes, vascular endothelia, mesothelia, and a subset of hematopoietic progenitors. Podocalyxin appears to complement CD34 as a useful cell surface marker for hemangioblasts, the common precursors of hematopoietic and endothelial cells.

SSEA-1 is expressed on the surface of preimplantation-stage murine embryos and has been found on the surface of teratocarcinoma stem cells, but not on their differentiated derivatives. SSEA-3 is synthesized during oogenesis and is present in the membranes of oocytes, zygotes and early cleavage-stage embryos. Biological roles of these carbohydrate-associated molecules have been suggested in controlling cell surface interactions during development.

Nucleostemin is a protein found in the nucleoli of embryonic stem cells, adult CNS stem cells, primitive cells in the bone marrow, and cancer cells. It is not in the differentiated cells of most adult tissues. It has been suggested to play a role in controlling the cell-cycle progression in stem cells and cancer cells.

The transcription factor OCT4, expressed in ES cells and germ cells, is strongly implicated in the process of maintaining as well as regaining stem-cell pluripotency and functions as a key regulator of mammalian germline development. It is expressed in totipotent embryonic stem and germ cells. A critical level of Oct-4 expression is required to sustain stem cell self-renewal and pluripotency.

Receptors for Vascular Endothelial Growth Factor (VEGF), VEGFB and PGF have a tyrosine-protein kinase activity. The VEGF-kinase ligand/receptor signaling system plays a key role in vascular development and regulation of vascular permeability.

Glial fibrillary acidic protein (GFAP) is a member of the class III intermediate filament protein family. It is heavily and specifically expressed in astrocytes and certain other astroglia in the central nervous system, in satellite cells in peripheral ganglia, and in non-myelinating Schwann cells in peripheral nerves. In addition neural stem cells frequently strongly express GFAP. Antibodies to GFAP are therefore very useful as markers of astrocytic cells. In addition many types of brain tumors, presumably derived from astrocytic cells, heavily express GFAP.

Microglia are the smallest glial cells that function similarly to macrophages in the blood system. They are the first immune defense in the CNS that seek and destroy damaged neurons, plaques, and infectious agents. When infections cross the blood-brain barrier, microglia cells act to increase inflammation in order to destroy the infectious agents before they cause damage to the brain and/or spinal cord. Due to their plasticity, microglia cells are present in various forms depending on their location in the body, such as ameboid, ramified, phagocytic, juxtavascular, and gitter cells.

Oligodendrocytes provide support to axons and produce insulating myelin sheaths. The CNS depends on this insulation to reduce ion leakage in order to increase overall nerve conduction speed. When myelin

sheaths are degenerated oligodendrocytes attempt to replace them, however, in multiple sclerosis patients oligodendrocytes themselves are destroyed thus compromising their ability to perform repairs.

PGP9.5/UCHL1 AntibodiesPeripheral Nerve Fiber Markers

Ubiquitin C-terminal hydrolase 1 (UCHL1) was originally identified as a major component of the neuronal cytoplasm from 2-dimensional gel analysis of brain tissues, and was given the name PGP9.5. The protein is extremely abundant representing 1-2% of total brain protein. It was later found that a ubiquitin C-terminal hydrolase enzyme activity was associated with the PGP9.5 protein, resulting in the renaming of PGP9.5 to ubiquitin C-terminal hydrolase 1.

Since UCHL1 is heavily expressed in neurons, antibodies to UCHL1 can be used to identify neurons in histological sections and in tissue culture. The great abundance of this protein in neurons means that it is released from neurons in large amounts following injury or degeneration, so the detection of UCHL1 in CSF and other bodily fluids can be used as a biomarker. UCHL1 was also discovered as a gene mutated in some rare famial forms of Parkinson’s disease. Interestingly, a common allelic variant of UCHL1, the S18Y polymorphism is actually protective against Parkinson’s disease.

Synapsin I plays a key role in synaptic plasticity in the brain. This effect is due in large part to the ability of the synapsins to regulate the availability of synaptic vesicles for release. In addition to its role in plasticity, the expression of synapsin I is a precise indicator of synapse formation. Thus Synapsin I immunocytochemistry provides a valuable tool for the study of synaptogenesis. The role of synapsin in synaptic plasticity and in synaptogensis is regulated by phosphorylation.

The centrosome is the main microtubule organizing center (MTOC) of the cell and acts as a regulator of cell-cycle progression. Centrosomes are present in higher eukaryotic cells, but not plant cells. The centrosome is composed of two centrioles surrounded by an amorphous mass of pericentriolar material

(PCM). The PCM contains proteins responsible for microtubule nucleation and anchoring. Each centriole is composed of nine triplet microtubule blades in a pinwheel structure. Cancer cells often have an abnormal number of centrosomes, which may be of biological significance.

Autophagy is an evolutionarily conserved process responsible for the bulk degradation of intracellular material. In autophagy, cytoplasmic components are engulfed by double-membrane-bound structures

(autophagosomes) and delivered to lysosomes/vacuoles for degradation. Recent reports indicate that autophagy plays a crucial role in many different pathologies.

LC3 is the only known mammalian protein identified that stably associates with the autophagosome membranes. LC3-I is cytosolic and LC3-II is membrane bound and enriched in the autophagic vacuole fraction. The detection of LC3-I to LC3-II conversion is a useful and sensitive marker for distinguishing autophagy in mammalian cells.

Pericentrin binds to calmodulin and is expressed in the centrosome. It is an integral component of the pericentriolar material (PCM). The protein contains a series of coiled-coil domains and a highly conserved PCM targeting motif called the PACT domain near its C-terminus. The protein interacts with gamma-tubulin and is likely important to normal functioning of the centrosomes, cytoskeleton, and cell-cycle progression.

Endosomes are membrane-bound vesicles that are formed by endocytosis. Endocytosis involves invagination of the plasma membrane to encircle a substance and budding to release the vesicle.
