The brain chemical missing in Parkinson's disease may have a hand in its own death. Dopamine, the neurotransmitter that helps keep body movements fluid, can kick off a toxic chain reaction that ultimately kills the nerve cells that make it, a new study suggests.

By studying lab dishes of human neurons derived from Parkinson's patients, researchers found that a harmful form of dopamine can inflict damage on cells in multiple ways. The finding also hints at a potential treatment for the estimated 10 million people worldwide with Parkinson's: Less cellular damage occurred when some of the neurons were treated early on with antioxidants, molecules that can scoop up harmful chemicals inside cells.

Study co-author Dimitri Krainc, a neurologist and neuroscientist at Northwestern University Feinberg School of Medicine in Chicago, and colleagues took skin biopsies from healthy people and people with one of two types of Parkinson's disease - inherited or spontaneously arising. The researchers then coaxed these skin cells into becoming dopamine-producing neurons. These cells were similar to those found in the substantia nigra, the movement-related region of the brain that degenerates in Parkinson's.

The researchers noticed some worrisome changes in the mitochondria of neurons carrying a mutation that causes the inherited form of Parkinson's after 70 days of growth. Levels of a harmful form of dopamine known as oxidized dopamine began rising in the mitochondria, reaching high levels by day 150. Neurons derived from people with the more common, sporadic form of Parkinson's showed a similar increase but later, beginning at day 150. Cells derived from healthy people did not accumulate oxidized dopamine. This dangerous form of dopamine seemed to kick off other types of cellular trouble. Defects in the cells' lysosomes, the cellular cleanup machines, soon followed. So did the accumulation of a protein called alpha-synuclein, which is known to play a big role in Parkinson's disease.

Those findings are “direct experimental evidence from human cells that the very chemical lost in Parkinson's disease contributes to its own demise,” says analytical neurochemist, Dominic Hare, Florey Institute of Neuroscience and Mental Health, Melbourne, Australia.

When researchers treated neurons carrying a mutation that causes inherited Parkinson's with several different types of antioxidants, the damage was lessened. To work in people, antioxidants would need to cross the blood-brain barrier, a difficult task, and reach the mitochondria in the brain. And this would need to happen early, probably even before symptoms appear, Krainc says.

HT: I was telling them about my experience on reaching the Nanda Devi basin. We ended up talking about the feeling of inner connection

although I knew we weren't allowed to climb the peak. Like you said, I was telling them mountains do offer moments of transcendence.

NR: I'm sure they do. See who has walked in with me. I'm sure you have been introduced to Stephen before, haven't you?

HT: Of course, we have.

NR: Stephen, I was touched by your story in Becoming a Mountain. You wanted to explore the tradition of Peter Matthiessen's The Snow Leopard,

on transcendence, didn't you?

SA: Yes. Like you started on an exploration of Shambhala, for me at that point in time it was a journey of healing, a pilgrimage of the soul, if

you like.

NR: Why 'at that point in time,' what was special about the time?

SA: I thought you'd have heard of the attack in 2008.

HT: Attack, what attack? SA: I thought you knew about it. It's a long story. I've written about it in the book.NR: Why don't you tell us about it Stephen?SA: Well, like I said it happened in July 2008, in our home. There were these four guys who crashed into our house early in the morning. We

were asleep, myself and my wife Ameeta.HT: In your house, in India?SA: Yes, in Mussoorie. They were armed with a country pistol and knives. They hit us, tied our wrists and legs, stabbed and threw us to the

ground trying to smother and kill us … shouting all the time “where is the money?” I kept shouting, 'there's no money … take whatever you

want, the TV, laptops, camera.' They wouldn't listen. 'Tell us where the money is or we will shoot your wife – madam ko goli mar dengey.' I

remember thinking it was strange that they called Ameeta 'madam' after they had beaten and stabbed her.NR: Good Lord! What a deadly nightmare!SA: It was. The whole thing comes back to me as if it were a cutaway shot in a black-and-white film. I remember glimpses of a cynical

amusement in the face of my attacker as though he took a sadistic pleasure in what he was doing. I was convinced I was going to die but

somehow didn't want to give up without some kind of resistance. Surprisingly I felt no pain even when his knife made big gashes in my flesh. It

Global NewsEbola Vaccinations Begin in Congo

Emergency teams responding to the ongoing Ebola outbreak in Congo began on May 21 inoculating those most at risk of contracting the virus: health workers and people who have come into contact with Ebola victims. It is the first real-world test for an experimental vaccine, rVSV-ZEBOV. In field trials in Guinea and Sierra Leone in 2015, this vaccine effectively protected people from Zaire ebolavirus — the same type of Ebola, which is now circulating in Congo.

In the latest outbreak, 51 people have developed hemorrhagic fever consistent with Ebola, and 27 died. Using a 'ring vaccination' strategy, healthcare workers are offering shots not just to the people who have had contact with Ebola victims, but also to a second ring of people who have had contact with the first group. In that way, WHO and its partners hope to disrupt the chain of transmission.

Merck, the company that makes the vaccine, has donated 8,640 doses which is more than enough for 50 rings of 150 people. Another 8,000 doses are expected to become available soon, according to WHO.
