Bio-Rad Laboratories

Prepare PBS. Cover moisture chamber/slide holder. Incubate substrate slides at room temperature for20 min (mouse) or 30 min (rat).

Prepare screening dilutions of the test sera as specified in the instruction manual.

Remove appropriate number of slides from freezer orrefrigerator and equilibrate to room temperature in foil bags (20 min). Bring all reagents to room temperature.

Gently rinse each slide with PBS buffer and remove excess buffer.

Place the slide(s) into a staining dish filled with PBS and incubate for 10 min.

Remove each slide or slide holder from staining dish and blot excess PBS from around wells using blotting strips. Do not touch strip to substrate.

Autoimmune IFA Procedure

Remove slides from foil bag and place in moisture chamber. Immediately add 50 µL of controls or diluted test sera to appropriate wells.

Place slide(s) in moisture chamber and immediately dispense 50 µL FITC conjugate to each well.

Cover moisture chamber or slide holder. Incubate 20 min (mouse) or 30 min (rat) at room temperature.

Rinse each slide briefly with a stream of PBS. Wash slides for 10 min in a staining dish filled with PBS.

Remove each slide or slide holder from PBS and drain briefly on a paper towel. Apply 4 ± 1 drop(s) mounting media perslide, making sure to cover all wells and add cover slip.

Examine the reactions on the slide under a fluorescent microscope.

Kallestad™ – the name you know, the quality you trustThe Bio-Rad rodent tissue slides are indirect fluorescent antibody reagents for the detection and semi-quantitation of human autoantibodies.

Our rodent tissue sections are highly sensitive substrates, which can be used to detectanti-keratin (AKA), anti-mitochondrial (AMA), anti-smooth muscle (ASMA), anti-parietal cell(APCA), anti-liver-kidney-microsomal (LKM), antibodies. These tissue sections alsominimize interference from blood group iso-antibodies while providing easily discerniblefluorescent patterns.

Clinical UtilityHigh AMA titers support the diagnosis of primary biliary cirrhosis. Low AMA titers may be detected in other liver disorders which include chronic active hepatitis and cryptogenic cirrhosis.

ASMA are present in high titers in the serum of 70% of patients with chronic activehepatitis. Low ASMA titers may be present in viral infections, malignancies and good health.

APCA occur in the serum of 90% of patients with autoimmune pernicious anemia. Withother clinical and laboratory data, a positive APCA result helps to distinguish autoimmunepernicious anemia from other megaloblastic anemias.

LKM are microsomal antibodies that exhibit characteristic staining of the cytoplasm ofliver cells and proximal tubules of the kidneys. They are primarily found in patients withautoimmune hepatitis.

AKA are highly specific for rheumatoid arthritis.
