Assay of paracetamol tablet Apparatus: Pistol and mortar, beaker, stirrer, volumetric flask, pipette, paracetamol tablets Standard solutions: 0.1N NaOH solution (4g/100ml of water) Stated potency: 500mg tablet Official limits: 95-105% Extinction value: 715 at 257nm Procedure: For assay of paracetamol. Take 20 tablets and weigh it. Grind the tablets and weight the amount of powder that contain 150mg of active ingredient and transfer it to 100ml volumetric flask. Now add 40-50ml of the NaOH 0.1N. Now add distilled water and shake it in a volumetric flask. This is the first dilution. Take 10ml of it and add in another volumetric flask. Add few drops of the NaOH solution and dilute to 100 ml. similarly prepare the third dilution. Adjust the wavelength of spectrophotometer at 257nm. Fill the solution in the cell from 3 rd dilution and measure absorbance at 257nm. Assay of paracetamol tablet

Apparatus:

Compare it with official limits. If range fall in official limits, the batch is passed and if not the batch is failed.

Observations and Calculations:

Paracetamol tablet have active ingredient= 500mg

Weight of 20 tablets= 10.1g

Weight of 1 tablet= 505mg

500mg of active ingredient is in =505mg

1mg active ingredient will be in = 505/500

150mg active ingredient will be in = (505/500)×150 = 151.5 mg

First dilution:

100ml of the solution will contain = 150mg

10ml of the solution will contain = 15mg

Second dilution:

100ml of the solution will contain = 15mg

10ml of the solution will contain = 1.5mg

Third dilution:

In 10ml of the solution, add few drops of the NaOH solution. This is third dilution.

Dilution factor: Solution in E1%1cm /solution in final dilution

DF= (100mg/100ml)÷(1.5mg/100ml)

= 666.7

Percentage purity= (Absorbance × Dilution factor) ×100/E1%1cm

Percentage purity= 1.03×666.7×100/715

=96.04%

Result:

The batch is passed because the percentage purity fall in official limits.
