Fast and easy start to purificationÄKTAexplorer systems are entirely controlled by UNICORN software, enabling a fast and easy start to purification. Users are able to create complete purification schemes for a range of biomolecules by using the method Wizard. This makes ÄKTAexplorer extremely easy to use and speeds up routine purifications. It reduces the need for expertise in programming, in chromatography, and in time-consuming literature searches, as all this knowledge is built into the system.

The UNICORN method Wizard provides a unique combination of flow configuration and method support for all chromatographic techniques including gel filtration, anion and cation exchange, hydrophobic interaction, affinity, and reversed phase chromatography. Using input parameters such as components in use, flow rate, sample volume, or gradient, the method Wizard builds up the method to suit your needs (Figs 3-5). The method Wizard eliminates time-consuming and repetitive method programming and uses column volume as a programming base for easy use with any size column.

Routine runs quickly optimizedTo quickly establish routine runs, the method Wizard supports programming of method scouting (Fig 6). These scouting methods make it easy to systematically vary run parameters in repeated, automated procedures, reducing optimization times.

Accurate result comparisonsAll data, including running conditions, are stored permanently by UNICORN within the run log (e.g., flow, pressure, UV, conductivity, temperature). This makes it easy to compare runs (e.g., Fig 7) and determine a cause for any deviations that may be seen.

Column knowledge built-inÄKTAexplorer ensures every column is run optimally and is protected from damaging events such as overpressure.

The user selects a prepacked column from the extensive UNICORN controlled column library (Fig 8) and ÄKTAexplorer automatically sets the optimal running parameters for that column (e.g., column volume, flow rate, and pressure limit) into the method. The user adjusts running conditions (if required) by simple method editing. New columns can be added to the library and will be run in the same secure way.

Fast, systematic method optimizationMethod optimization and process development on chromatographic systems often involves systematic variation of key run parameters, such as media selection, binding and elution buffers, or sample loading flow rates. UNICORN control system includes scouting schemes within each general method that eliminate the need for creating new methods for each set of parameters. These scouting schemes make it easy to systematically vary any parameter in repeated automated runs, without creating a large number of individual methods (Fig 6).

The method wizard supports the most common scouting procedures:

• Automatic media screening (Fig 9)

• Automatic eluent pH scouting for ion exchange chromatography (Fig 14)

No manual titration of buffersThe BufferPrep function increases speed and the reliability of results by eliminating the time-consuming manual buffer preparation and titration usually needed for every pH change in chromatography.

The user selects the buffers from the buffer library and defines the pH required (Fig 10). BufferPrep prepares correct buffer pH on-line from four stock solutions (e.g., water, salt, buffer, acid/base) and controls the pH throughout the run, automatically compensating for changes in temperature and salt concentration. New buffer substances can be added to the buffer library and used by BufferPrep to create the required pH elution conditions.

The accuracy of pH is crucial in many separations (Fig 11). The accuracy of BufferPrep greatly increases the reproducibility and reliability of results from day to day by eliminating and correcting errors that occur when producing buffers manually.

BufferPrep is used for pH scouting in rapid method optimization. The method wizard makes programming of automatic pH scouting for ion exchange chromatography easy and safe. Figure 12 shows BufferPrep used on ÄKTAexplorer 100 at 60 ml/min to test seven pH conditions to find the optimal pH for binding and elution of a pure Fab fragment.

Flexibility and safety from laboratory to productionEasy system modificationsNew functions to enhance system capabilities or create new system configurations are easily added. Functions, consisting of hardware components and software instructions, are selected, installed, and implemented by the user. UNICORN controls and monitors the new functions in the same way as all other ÄKTAdesign components.

Fast, trouble-free transfer of optimized methods to production scaleUNICORN control system is used at operational scales from the laboratory bench to full production. UNICORN combines the flexibility needed for method and process development with the stringent requirements for commercial manufacture of biopharmaceutical products on production chromatography systems.

The same user interface and methods used to optimize a purification are applied in commercial manufacture with larger systems. This common control platform means quick and simple transfer from one stage of a project to the next. Figure 13 shows a 10-fold scale-up of a Phenyl Sepharose™ 6 Fast Flow column during process development of a Fab fraction purification, using an ÄKTAexplorer 100 and the same method template.

to use and gives a complete overview of operations. With defined access rights, operators connect to and control systems from any remote control or local PC. The user interface is exactly the same. Results can be automatically saved on a server and evaluation and generation of reports can be made locally or at remote PCs.

ÄKTAdesign serviceRegular, planned maintenance increases productivity by keeping a system in top working condition and minimizing downtime. A service agreement lets users budget for all service and maintenance costs, giving full cost control and eliminating unexpected expenses. Service agreements greatly simplify compliance with regulatory and quality demands.

Scheduled preventive maintenance carried out by qualified engineers and the built-in service diagnostics of ÄKTAdesign systems provides users with support to meet the requirements of Good Laboratory Practice (GLP). All systems are supplied assembled and tested. Requiring minimal bench space, each system is mounted on a swivel platform for easy access to every side. Columns are snapped in place on the outside of the system, either individually or as groups held in column holders.

System componentsÄKTAexplorer systems have three main instrument components, a number of ancillary items, the option of a fraction collector and an autosampler (Fig 15). All instrument settings and functions are under direct control of UNICORN.

At every start-up, each instrument automatically performs a self-diagnostic test and calibrates appropriate settings. This ensures consistent performance over the long-term.

All wetted materials are inert, fully biocompatible, and resistant to all eluents commonly used in liquid chromatography, ensuring high recovery and no loss of biological activity.

Pumps provide reliable resultsThe main challenges of a chromatography system pump are to achieve accurate gradient formation over a broad flow rate range and avoid introducing air into the system. ÄKTAexplorer systems are equipped with either Pump P-901, which enables a wide flow rate range for both laboratory- and pilot-scale applications without changing pump modules or pump heads, or with Pump P-903, which operates at the lower flow rates and higher pressure required for running high-resolution analyses at laboratory scale. Figure 16 shows the flow rates and pressure ranges of the two pumps.

Monitor pH/C-900Monitor pH/C-900 is a combined monitor for accurate, online measurement of pH, conductivity, and temperature. The monitor records real-time information about chromatographic conditions. Monitor pH/C-900 provides an accurate response, coupled with high precision over a wide measuring range. This makes Monitor pH/C-900 an excellent choice for use in all chromatography techniques, from reversed phase chromatography with very low conductivity eluents to hydrophobic interaction chromatography in high salt solutions.

Monitor pH/C-900 consists of a control unit, an optional flow cell for conductivity and temperature, an optional flow cell with a holder for the pH electrode, and the pH electrode.

Monitor UV-900Monitor UV-900 is a high sensitivity, multi-wavelength UV-Vis monitor, recording up to three wavelengths simultaneously in the range 190 to 700 nm, which is the optimal detection range. The use of advanced fiber optics together with the unique flow cell design ensures a high signal-to-noise ratio for maximum performance. Maintenance costs are minimized by use of a unique Xenon flash lamp, giving a significant increase in lamp lifetime.

ÄKTAexplorer 100 chromatography systems include a UV-900 monitor with a 2-mm pathlength flow cell (internal volume 2 µl). The flow cell in ÄKTAexplorer 10 chromatography systems has a pathlength of 10 mm (internal volume 9 µl).

Ancillary equipmentStandardSample Pump P-960

Pump P-960, together with an IV-908 valve, is used for automatic multiple sample injection. The flow rate range (up to 50 ml/min), low pulsation, and pressure specification (up to 1 MPa) allow rapid sample application directly onto a column or via a Superloop. Large sample volumes can be applied smoothly and safely under the control of UNICORN. This function is standard in ÄKTAexplorer 100, ÄKTAexplorer 100 Air, and ÄKTAexplorer 10 S and optional in other systems.

Valves

Seven-port injection valves, INV-907, are used for sample application and reverse flow elution.

Eight-port fractionation valves are used for buffer selection (IV-908), automatic sample application, column switching and flow diversion to fraction collector, large fraction collection, or waste (PV-908).

Two switch valves work together with the P-900 pump in BufferPrep mode.

Mixer M-925

Mixer M-925 is a dynamic, single chamber mixer powered and controlled from Pump P-900. For optimal mixing in the entire flow rate range of each ÄKTAexplorer system, interchangeable mixing chambers are supplied. ÄKTAexplorer 100 includes 2, 5, and 12-ml chambers while ÄKTAexplorer 10 has 0.6 and 2-ml chambers.

OptionalFraction collector

Frac-950 (Fig 15) prevents sample loss across the entire flow rate range of ÄKTAdesign systems, from 0.001 ml/min to 100 ml/min, and the highest flexibility in the choice of collection mode. Fractions are collected in microplates (over 384 fractions, up to 2.5 ml per fraction), a variety of test tube sizes, or in 20 large vessels (up to 250 ml per fraction) or even funnels for unlimited fraction size. Collection is in volume or time mode and different fraction sizes can be collected during different stages of a separation.

Autosamplers

Autosamplers give you more time to work on other projects during the day, and increase throughput by enabling overnight purification runs. Depending on your needs for sample injection, Autosampler A-900 and A-905 for ÄKTAdesign systems offer a variety of alternatives.These autosamplers offer different balances between reproducibility and accuracy and allow flexible use of vials and microplates. Almost every type of vial with an o.d. of 12 mm or 7 mm can be used on Autosampler A-900. Autosampler A-905 handles standard and deep-well 96-well microplates, as well as 384 microvials. Sample loss is kept to a minimum or eliminated by the novel method of sample withdrawal. The Peltier cooling is incorporated in both autosamplers and effectively protects heat–sensitive biomolecules during sampling. Autosampler A-900 and Autosampler A-905 are directly controlled by UNICORN software.

Air sensors

Controlled through UNICORN, air sensors are used together with the UNICORN WATCH programming function to increase control over programmed events. For example, UNICORN is programmed to ensure that chromatographic elution begins only when completion of sample application has been detected by the air sensor. Large, variable sample volumes are applied with no risk of air entering the system.

Flow control

Motorized valves INV-907, IV-908, or PV-908 can be added to allow additional programmable functions such as automated reverse flow through a column or flow diversion.

AD-900 Analog/Digital Converter

AD-900 Converter is an analog/digital converter for connecting external instruments to UNICORN control software. The AD-900 Converter allows easier monitoring of signals received as voltage inputs from external sources such as fluorescence detectors, radioactivity monitors, refractive index monitors, or diode arrays.
