Canadian Embryo Transfer Association / Association Canadienne de Transfer d’Embryons 2020 – VOLUME II NEWSLETTER / BULLETIN Dear CETA/ACTE Members, As I am writing this, the summer is in full swing. In Ontario, Québec and the Maritimes it has been a very hot and dry summer so far as opposed to Western Canada who has experienced a cooler and wet summer. As practitioners, we are always very busy; summer is a good time to spend some time with the family and to take some time for yourselves such as going on hikes, cycling, rollerblading, swimming, etc. I promised myself I would not talk about the Covid-19 pandemic! It is very hard not to do so as it has made us cancel the AETA & CETA/ACTE joint convention in Madison. When Covid hit us in March, we all thought that it would be over by the fall but it looks like it will be here for a while. The good news in all of that, if there is one, is that AETA/CETA will get the deposit back from the venue and there will be no penalty for cancelling. AETA has been busy in trying to figure out how to deliver a virtual conference. More details will be coming out very soon as you will have to register for the virtual joint convention. It will be a totally new concept. I strongly believe that we can deliver good quality CE with such a format. As you are aware, we had started the process of finding a replacement for Karen. I am happy to report that we have hired the new secretary-manager for CETA/ACTE. Carole Ness of Howick, Québec will be replacing Karen in the new year. She brings everything we were looking for; she is a detailed focus manager with loads of experience in multi-tasking, office management, book keeping and computer knowledge. As the CETA/ACTE office will be moving to Québec in the new year, the Articles for CETA/ACTE filed with the Canada Not-for-Profit Corporations Act will need to be changed, as well as a couple of By-law changes. These will be presented at the AGM which will be held virtually this year on October 7. This is my last president’s message. Dear CETA/ACTE Members,

As I am writing this, the summer is in full swing. In Ontario, Québec and the Maritimes it has been a very hot and dry summer so far as opposed to Western Canada who has experienced a cooler and wet summer. As practitioners, we are always very busy; summer is a good time to spend some time with the family and to take some time for yourselves such as going on hikes, cycling, rollerblading, swimming, etc.

I promised myself I would not talk about the Covid-19 pandemic! It is very hard not to do so as it has made us cancel the AETA & CETA/ACTE joint convention in Madison. When Covid hit us in March, we all thought that it would be over by the fall but it looks like it will be here for a while. MEMBERSHIP NOTICES

From the CETA/ACTE Office:

2019-2020 Annual Report: The Annual Report will be emailed to members. The report will include draft financial statements, committee reports, proposed amendments to the CETA/ACTE By-laws and proposed amendments to ‘Articles’. Please print a copy for the AGM held virtually on Wednesday, Oct 7, 2020.

CETA/ACTE Certified Practitioners: A CE/Embryo Summary was emailed to each certified member the beginning of July, please keep this information on file for easy reference regarding:

CE credits: the number you currently have on file and the number of credits needed for re-certification.

Number of embryos processed: the number you currently have on file and the number still required for re-certification.

Also included on your summary is the date that you will apply for re-certification and the date the requirements must be completed for re-certification.

APPLYING FOR CETA/ACTE CERTIFICATION:

To apply for CETA/ACTE Certification an application and the required information must be submitted to the CETA/ACTE office by May 1 (late applications will be accepted up to June 30th and levied a late penalty fee). Once your application and all information are received it will be reviewed by the Certification Committee to determine if you have met the requirements and are eligible to write the CETA/ACTE Certification Exam. Please note, once successful with the written exam, a mandatory site visit (practical exam) is required. If you are interested in applying for certification, please note the following: Please contact the CETA/ACTE office and request an

application to apply for certification, please indicate the species and technique(s).

The certification exam is only offered at the time of the annual convention.

NOTE: If you applied for re-certification this year, all CE and Embryo numbers included on your summary will be used for re-certification and cannot be carried over to your new 3-year certification term.

If you did not receive your CE/Embryo summary, please contact the CETA/ACTE office immediately.

CETA/ACTE Web site: The ‘ET Activity in Canada for 2019’ has been posted on the web site, on the ‘ET Activity in Canada’ page.

A minimum of 10 hours of continuing education related to embryo transfer within the past 2 years, is required when applying for certification.

Dates and location of future CETA/ACTE Certification Exams include:

October 2021, Vancouver, British Columbia, Canada October 2022, Louisville, Kentucky, USA October 2023, Orlando, Florida, USA October 2024, Toronto, Ontario, Canada

CETA/ACTE CERTIFICATION The CETA/ACTE Certification program is often used as a model by

other embryo transfer associations in the development of their certification programs.

CETA/ACTE Certification is a required and first step in the Canadian Food Inspection Agency (CFIA) Embryo Export Approval Program (EEAP) in order to export embryos from Canada.

CETA/ACTE Certification is also a measure of excellence and is used by practitioners to promote their expertise in the field of embryo transfer.

In Memoriam: Dr. Vlad Pawlyshyn Received from Dr. Reuben Mapletoft:

It is with sadness that we inform membership of the death of Dr. Vlad Pawlyshyn. Vlad passed away peacefully, at home, in Martensville, SK on June 9, 2020.

After graduation from the University of Saskatchewan in Veterinary Medicine (WCVM) in 1975, Dr. Pawlyshyn went to private practice in Langenburg, SK, and then returned to the WCVM in 1980, completing a Master of Veterinary Science under the supervision of Dr. Bill Cates. Vlad was an integral member of the Reproduction Research Trust Unit during the pioneering days of on farm bovine embryo transfer conducted by the WCVM, travelling throughout Saskatchewan, Manitoba and Alberta to provide embryo transfer services for cattle breeders. In 1985, Vlad established a reproduction practice, Trans Tech Genetics, serving clients across Western Canada. Throughout his career, he maintained a close relationship with the WCVM Theriogenology group, providing embryo transfer services for research projects, teaching practitioners in the WCVM Embryo Transfer workshops and training countless graduate students and clinical residents in Special Topics courses in embryo transfer. Vlad was one of the first certified embryo transfer practitioners in Canada, certified by the Canadian Embryo Transfer Association in 1993. He travelled to Argentina to transfer embryos on more than one occasion. Dr. Pawlyshyn was a past board member and president of the Canadian Embryo Transfer Association. He was also a regular attendee at IETS meetings, regardless of where they were held. Dr. Pawlyshyn had many friends throughout the embryo transfer industry and in the wider community of veterinarians, and he will be dearly missed.

AETA & CETA/ACTE Joint Virtual Convention

October 6 - 7, 2020

Registration is open for the 2020 AETA & CETA/ACTE Joint Virtual Convention. The convention will take place virtually from October 6-7 and will feature two types of sessions: keynote addresses and prerecorded sessions. Keynote addresses will be live sessions via Zoom, where participants will see the content and interact with the speakers in real time. Once the keynote addresses have concluded, a link to the content will be posted. Prerecorded sessions will be made available via a link on the AETA convention page at 12:01 am (CDT) on Tuesday, October 6. During the times on the schedule, speakers will be available to chat via Zoom about their presentations and answer any questions you may have. All sessions will have a quiz that participants must participate in and pass with a score of at least 70% to receive continuing education (CE) credits. Please note that speakers will only be available for discussion during their scheduled time. Participants who register before October 6th can view sessions and take the accompanying RACE quizzes until December 31st. You must register before October 6th to get access to the meeting content. Bovine oviductal and uterine fluid support in vitro embryo development

The study investigated the effect of oviductal fluid (OF) and/or uterine fluid (UF) supplementation on in vitro embryo development and quality. A lot of advancement has been achieved in the last decade in in vitro embryo production (IVP). However, the quality of IVP blastocysts are still considered lower than in vivo produced blastocysts. Their morphology, metabolism and cryotolerance are different. Modification of the in vitro culture environment can improve blastocyst production and quality. Supplement like fetal calf serum (FCS) or bovine serum albumin (BSA) are commonly used in culture media. FCS, despite the fact that it accelerates embryo development, have been associated with a lower blastocyst quality and fetal overgrowth at birth. Lots of studies have been done on different oviductal molecules but there is a limited number of studies regarding OF or UF supplementation during IVC.

After collecting cumulus oocytes-complexes (COCs) from slaughterhouse ovaries from mature heifers, standard in vitro maturation and in vitro fertilization was achieved. Presumptives zygotes were splitted 18 to 20 hours after fertilization in five different groups for IVC: 1- Synthetic oviduct fluid (SOF) supplemented with 5% FCS (SOF+FCS) 2- SOF supplemented with bovine serum albumin (SOF+BSA) 3- SOF with 1,25% OF from day 1 to day 4 (OF group) 4- SOF with 1,25% OF from day 1 to day 4 followed by 1,25% UF from day 4 to day 9 (OF+UF group) and 5- SOF with 1,25% UF from day 4 to day 9 only (UF group). The ability of the blastocyst to tolerate cryopreservation was used as an indicator of quality. Gene expression analysis was also performed on the blastocyst from the different groups, as well as evaluation of the reactive oxygen species (ROX) quantification via fluorescence. Results

On the embryo development side, no difference in the cleavage rate were noted between all groups. Blastocyst production was higher at Day 7 in the SOF+FCS group compared to SOF+BSA and OF groups, but similar to OF+ UF and UF groups. At Day 8 and 9, no differences were observed on the blastocyst rate of all groups.

The blastocyst survival rate 24h after warming was not different between groups, ranging from 59.0% to 74.2. After 48h, survival rates of blastocysts obtained from culture with OF+UF and OF were similar to those of the SOF+BSA group, and significantly higher than those of the SOF+FCS group.

In relation to the gene expression, expression of the epigenetic control genes DNMT3A and IGF2R (genes important in embryo methylation) were upregulated in the OF compared to the SOF+FCS. Also, oxidative stress regulation genes was downregulated in the UF and OF+UF groups compared to SOF+BSA and OF groups.

The table 2 represent the relative reactive oxygen species (ROS) between groups.

Relative ROS fluorescence intensity was significantly lower in blastocysts from the OF+UF, OF and UF groups compared with blastocysts from the SOF+FCS groups. Also, ROS levels were significantly lower in OF+UF groups compared to SOF+BSA.

Discussion

The main finding of the present study is that a low concentration of OF and/or UP supports embryo development and improves quality. OF seems to provide control in embryo methylation, whereas UF seems to provide antioxidant activity.

Despite the successful improvement of IVP in cattle, there is evidence that post fertilisation culture environment have major impact on blastocyst quality. There is also evidence of a marked contrast in the pattern of gene expression related to lipid metabolism and oxidative stress response when comparing in vitro produced blastocyst to those produced in vivo, with an impact on embryo tolerance to cryopreservation. In this study, culturing embryo with OF seems to support embryo development, and similar effects were observed in other studies. However, this study suggest that alternating OF with UF during the culture period might mimic more accurately the physiological conditions in vivo. The period studied is a time of major developmental events occurring in the embryo, such as first cleavage, embryonic genome activation, morula compaction, blastocyst formation and hatching. As those events appear to be sensitive to culture conditions, there might be a positive effect of adapting the culture condition during the IVC period instead of using a static IVC.

In vitro production and manipulation of embryos increase reactive oxidative species generation. This is related with impaired development of embryos. Conclusion

In conclusion, low concentrations of OF and UF with serum-free culture media supports embryo development, blastocyst quality and cryotolerance. This evidence might lead to the discovery of specific proteins important for embryo development and help improve the success of IVP. ֎
