Module Connections and Inputs

TheOrionVERSASTARmetercanacceptOrioncombinationISEelectrodesaswellasseparatereferenceandsensing

half-cellelectrodes.ThisallowsforawiderISEmeasurementoffering.Half-cellelectrodesseparatetheionssensing

fromthereferenceelectrode.Fordifficultsamples,theflexibilityofusingalternatefillsolutionsmaybeanadvantage

to researchers.

1. PreparethepHorISEelectrodeandATCprobeforuseasinstructedintheelectrodeuserguides.

The pin tip reference connector is on the lowest panel of the pH/ISE module.

2. PlacethepHorISE,ATCprobeandOrionStarstirrerprobe(cat.no.096019,optional)intothe

electrode arm.

3. AttachthepHorISEBNCandATCprobetothepH/ISEmetermodule.LowerISEandATCinto

solutionforconditioningifrequired.Ifusingthestirrerprobe,attachprobetothemeter,placeintothe

electrodearmandassigntheprobetothechannelinthesetupmenuforinstrumentsettings.

4. Preparefreshstandardsdailyforcalibration.ForISEtesting,standardsshouldbeatleastonedecade

(10times)apart.Thestandardsshouldexceedtheexpectedminimumandmaximumsamplerange.

Serialdilutionisrecommendedusingcalibratedpipettestoaccuratelypreparecalibrationstandards

from a stock solution.

5. Gatherreagents,rinsewater,andothersupplies.

ForISEtesting,ifISAwillbeaddedtothesample,itneedstobeaddedtoallthestandardspriorto

calibrationtoensureaconsistentdilutionfactor.

Module Maintenance

Forroutinemaintenance,dustandwipethemodulewithadampcloth.Ifnecessary,warmwaterormildwater-based

detergentcanbeused.Maintenancecanbeperformedonadaily,weeklyormonthlybasis,asrequiredbyoperating

environment.Immediatelyremoveanyspilledsubstanceusingthepropercleaningprocedureforthatspilltype.

usethe“X”electrodewiththevalence(+1,+2,-1,or-2)matchingyourelectrodefor

accurate results.

Read Type Information

Therearethreemainreadtypesavailableasshowninthesetupmenus.

• Auto-Read – Themeterwilldisplaythemeasurementasitstabilizesandlockandholdthe

measurementwhenitisstable(TheARiconwilllockonthescreen.)Oncethemeasurementislocked,

themeterwillautomaticallyexportthemeasurementtothedatalog,ifthedatalogisenabledin

thesetupmenu,andtoaprinterorcomputer,ifaprinterorcomputerisconnectedtothemeterand

enabledinthesetupmenu.Totakeanewmeasurement,pressmeasure (esc)/1.

• Timed – Themeterwillreadanddisplayameasurementatthesettimeintervalthatisprogrammed

bytheoperator.Therearetwooptions:

1. Bychecking/selectingthe"Interval"box,atthesettimeinterval,themeterwillautomatically

exportthemeasurementtothedatalog,ifthedatalogisenabledinthesetupmenu,andto

aprinterorcomputer,ifaprinterorcomputerisconnectedtothemeterandenabledinthe

setupmenu.Ifselected,enterthetimeintervalvalueinahours:minutes:secondsformatusing

the numeric keypad and press f3 (select) key.

2. Ifthe"Interval"boxisunchecked/notselected,asingletimedmeasurementwillbetaken.

Themeterwillapplythesamesavingandexportingconditionsaswiththesettimeintervalto

recordandexportdata,butlockafteronetimedintervalhasexpired.Pressmeasure (esc)/1 to

• Continuous – The meter will continuously measure and update the display. This read type is useful

whenperforminganexperimentthatrequirescontinuousmeasurementstobetaken.Pressthe

log/print/0printkeytoexportthemeasurementtothedatalog,ifthedatalogisenabledinthesetup

menu,andtoaprinterorcomputer,ifaprinterorcomputerisconnectedtothemeterandenabledin

the setup.

Isopotential Value

Theslopeofallelectrodeschangeswithtemperature,basedontheNerstequation.Theslopecurvesatthedifferent

temperaturesintersectattheisopotentialvalue,whichistheconcentrationatwhichthepotentialoftheelectrodedoes

notvarywithtemperature.ForglasspHelectrodes,thisvalueis7.00.Forotherionselectiveelectrodes(ISEs),ifthe

isopotentialvalueisknownorcanbemeasuredexperimentally,adjustingthisvaluemakestemperaturecompensation

foranISEpossible.Temperaturecompensationcanbeperformedforoneandtwopointcalibrations.

Linear Regression Information

Thedefaultisoffwhichusesthetraditional,segmentedapproach.Thesegmentedapproachisthemostaccurateand

precise.MoreinformationcanbefoundintheappendixsectionofthisreferenceguideundertheMultipointCalibration

Curve Feature.

Selecting“on”forthisoptionresultsintheuseofasinglebestfitlineforthecalibrationcurve,andcanbebeneficialif

pHmeasurementsvarygreatly.

Low Level Stability

ThisfeatureadjuststhetimingforthecalibrationpointsforlowerlevelISEstandards.

Setup Menu for Temperature

AllofthetemperaturemeasurementsettingsarefoundinTemperatureSetupmenuforeachmeasurement.

*Thisnumberwillchangetomatchthesetupmenuchannel.

Temperatureprobescanbesetupinthemeasurementmodeofthesampleyouaremeasuring.Ch-1ATCallowsthe

temperatureprobetobeusedinmorethanonechannelfortemperature-compensatedmeasurements,suchaswhen

apHelectrodeanda2-cellconductivitycellareinthesamemeasurementsample.TemperatureisacriticalpHand

conductivitymeasurementasthemeasurementvaluesrequireatemperaturecorrectiontodeterminethereference

value,often25ºCintheAmericaand20ºCinEurope.

concentrationfirst.Alwaysusefreshstandards.Selectstandardsthatbracketthesampleconcentrationandarea

decadeapart.Preparetheionselectiveelectrodeaccordingtotheelectrodeuseguide.

1. Presschannel key until ISE channel is displayed. Press Cal (f1).Followthescreenpromptsstarting

with the lowest concentration.

2. PressEdit (f3)tothecorrectconcentrationvalueofastandardifitisnotautomaticallyrecognized.

3. Afterthefirstdatapointhasbeenaccepted,pressAccept (2) tocontinuecalibration.

The meter then offers options of Esc (f1)toleavecalibration,Next (f2)measurenextcalibration

point,orCal Done (f3).

4. TheoptionsafterCalDoneispressedarePrint (f1),Cal Edit (f2) or Slope Edit (f3).

5. TheCalibrationreporttableisdisplayedwiththeconcentration,mVreading,andTemperatureand

averageslopewhichisupdatedwitheachsuccessivepointmeasured.

6. Fromthisscreenthenavigationoptionsare Print (f1),Cal Edit (f2) or Slope Edit (f3).Printexports

datatoprinterorcomputer,Caleditallowsconcentrationtobecorrectedpriortothecompletionof

thecalibrationbyDelete (f1) or ReMeasure (f3).

7. PressingtheSlopeEditchangesthescreentitletoSinglePointCal–SlopeEdit.Thisallowsthe

currentslopelinetobeupdatedwiththepresentelectrodeperformance.

8. Press Cal Done (f1)whencompleted,pressviewgraph,savethecalibrationinCalLog.

Known Addition and Subtraction

TheOrionVERSASTARmeterhaspresetmethodsforsingleknownaddition,singleknownsubtraction,doubleknown

additionanddoubleknownsubtraction.

• Whenperformingincrementaltechniques,thesampleconcentrationmustbeknownwithinan

orderofmagnitudesotheincrementmaybecorrectlychosenandthesampleandstandardmustbe

volumetrically determined.

• Itisveryimportantthatanycomplexingagentispresentinexcess(atleast50to100times)ornot

atall.Theratiooffreetocomplexedionsmustremainconstantovertheaddition.Anindicationthat

acomplexingagentispresent,butnotingreatenoughquantitytomaintainaconstantfreeionto

complexedionratio,isanabnormallyhighorlowslope.Ifthisisasuspectedproblem,addanexcess

amountofcomplexingordecomplexingagenttotheoriginalsolutioninthebeaker.

• Electrodeinterferencesshouldbeataminimum,sincetheeffectoftheinterferencemightchange

astheconcentrationoftheionofinterestchanges.Thissituationmayalsoresultinhighorlow

electrodeslopes.Consulttheelectrodeuserguideforinformationonreducingoreliminatingsome

electrode interferences.

• Knownadditionisausefulmethodformeasuringdilutesamples,occasionalsamplesorsamplesthat

containanexcessofcomplexingagent.Knownadditioncanalsobeusedtoverifytheresultsofa

direct measurement.

• Knownsubtractionisusefulwhenmeasuringionsforwhichstablestandardsdonotexist.Known

subtractioncanalsobeusedasaquickversionofatitration.Whenperformingaknownsubtraction,

itisnecessarytoknowthestoichiometricratiobetweenthestandardandsample.

Testing a pH Electrode for Automatic Buffer Recognition

ToverifythattherawmVreadingofthepHelectrodeinuseis±30mVfromthetheoreticalmVreadingofthebuffer,andthereforeverifythatthepHelectrodeinuseiscapableofperformingautomaticbufferrecognition,performthefollowingprocedure.

1. SetthemeasurementmodetomV.PreparethepHelectrodeaccordingtotheinstructionsinthepHelectrodeuserguide.

2. RinsethepHelectrodewithdistilledwater,blotitdrywithalint-freetissueandplacethepHelectrodeintoapH4.01bufferatapproximately25°C.

3. WaitforthemeasurementtostabilizeandrecordthemVvalueofthepH4.01bufferwhenthemeterindicatesthatthemeasurementisstable.ThereadtypeselectedinthesetupmenuwilldeterminehowthemVmeasurementsaredisplayedbythemeter.

4. RemovethepHelectrodefromthepH4.01buffer.

5. RinsethepHelectrodewithdistilledwater,blotitdrywithalint-freetissueandplacethepHelectrodeintoapH7.00bufferatapproximately25°C.

6. WaitforthemeasurementtostabilizeandrecordthemVvalueofthepH7.00bufferwhenthemeterindicatesthatthemeasurementisstable.

7. ThemVreadingofthepHelectrodeinpH4.01buffershouldbeintherangeof+207to+147mVandthemVreadingofthepHelectrodeinpH7.00buffershouldbeintherangeof-30to+30mV.IfthemVreadingsofthepHelectrodeareinthecorrectranges,thepHelectrodeiscapableofperformingautomaticbufferrecognition.IfthemVreadingsofthepHelectrodearenotinthecorrectranges,thevaluesofthepHbufferswillneedtobemanuallyenteredduringapHcalibration.

Multipoint Calibration Curve Feature

TheOrionVERSASTARMeterwithpH/ISE/Temperaturemodulesystemsutilizeapoint-to-pointschemeformultipointcalibrationsinthepHandISEmodes.Inthistechnique,straightlinesegmentsaredrawnbetweeneachsuccessivepairofcalibrationpoints.SeparateslopeandE0valuesarecalculatedusingtheadjacentpairofdatapoints.Forexample,inafivepointcalibration,fourslopesandfiveE0valuesarecalculatedtocorrespondtothefourlinesegmentsthatconnectthedata.pHorconcentrationvaluesforsamplesarecalculatedaccordingtothesegmentthattheirpotentialvaluesfallinto.Thoseaboveorbelowtherangeofcalibrationarecalculatedbyextrapolationfromthetoporbottommostsegment.

Thedisplayedslopeisanaverageslopeoftheseparatesegments.Theslopemaybeoutsidetherangenormallyexpectedforanelectrode.Forexample,iftheslopeofonesegmentislow,thentheaveragewillbelowerthantheoretical,eventhoughtheresponsemaybeNernstianovermostoftherange.

Automatic Blank (Non-linear) Correction Feature

TheOrionVERSASTARMeterwithpH/ISE/Temperaturemodulesystemshaveafeatureknownasautomaticblankcorrection.Inthistechnique,themeterdecideswhetherblankcorrectionisthebestmeasurementstrategybyanalyzingtheelectroderesponseduringamultipointcalibration.Aseparateblankdoesnothavetoberun.Graphically,automaticblankcorrectionisequivalenttodrawingasmoothcurvethroughthelowestthreepointsofthemultipointcalibrationandextrapolatingtozeroconcentrationbasedontheassumptionofNernstianelectrodebehavior.

Multipointcalibrationsatthelowerlimitofdetectionaredesirablewhentheresponseofanionselectiveelectrodeisnon-linearandcannotbecharacterizedwithaoneortwopointcalibration.Thisisusuallyseenasalowelectrodeslope.Generally,theelectrodeisbehavinginaNernstianmannerbuttheeffectofablankisbeingobserved.Seethefigurebelow.

Thisblankmaybeatruereagentblank,tracesofanalyteioninthereagentsoritmaybethe“mud”valueoftheelectrode.Itcouldalsobeaninterferenceinthereagentsthatbecomesapparentatlowlevelsofanalyteion,oritcouldbeanycombinationoftheseeffects.TheexpandedversionoftheNernstequationtraditionallyusedforblankcorrectionis as follows:

Inamulti-pointcalibration,asetofequationsisgeneratedandtherelationshipbetweenthemevaluated.Forexampletheequationsgeneratedinathreepointcalibrationwouldbeasfollows:

E3 = Eo + S x log (C3 + b)

ThemeterevaluatestherelationshipsbetweenthethreepotentialsE1,E2andE3andthethreeconcentrationsC1,C2 andC3.Iftherelationshipsindicatethatblankcorrectionisdesired,ablankwillautomaticallybecalculatedandthe non-linearitywillbecorrectedforinaNernstianmanner.Iftheappropriateconditionsarenotmet,theblankissetatzeroandeachsegmentofthemultipointcalibrationistreatedindependently.

Whenallthreeofthefollowingconditionsaremet,blankcorrectionisinvoked.

1. Theconcentrationofthefirststandardiszero,ortheslopeoftheelectrodebetweenthefirstandsecondstandardsislessthantheslopebetweenthesecondandthird.

2. Potentialdifferencesbetweenpointsaresignificant.Forexample,E3-E1>10mV

3. Theblankcorrectionalgorithmconvergesatreasonableblankandslopevalues.Conditionsinsteps1and2preventfailuretoconvergeinmostsituations.However,theslopewillattainanyvaluenecessaryforconvergencetoacalculatedblankvalueof3xC3.

Whenblankcorrectionisimplemented,theslopevaluefortheelectrodemaybeoutsidetherangeofvaluesnormallyconsideredacceptableduringanordinarycalibration.Forbestresults,calibrationstandardsshouldbecloseinrangetotheexpectedsampleconcentrationsandshouldbrackettheexpectedsampleconcentration.Whenconditions1,2and3arenotmet,thecalibrationdataishandledbythemultipointcalibrationmethoddescribedearlier.

Incalibrationswithmorethanthreepoints,acombinationofmethodsisutilized.Automaticblankcorrectionisusedifthelowestthreepointssatisfythecriteriaandmultipointcalibrationisusedfortheotherpoints.Theslopedisplayedaftercalibrationisanaverageoftheslopecalculatedintheautomaticblankcorrectionalgorithmandtheslope(s)foreachadditionalsegment.

Determining an Isopotential Value

1. Prepareseveralstandardswithconcentrationrangesoverthemeasuringrangeoftheelectrode.

2. Measurethemillivoltvalueofthestandardsatroomtemperature,about20°Cto25°C.

3. Measurethemillivoltvalueofeachstandardat75°C.

4. Measurethemillivoltvalueofeachstandardat10°C.

5. Onsemi-logarithmicgraphpaper,plottheconcentrationvaluesonthelogaxisversusthemillivoltvaluesonthelinearaxis.

6. Thelineswillintersectattheisopotentialvalue.Readtheconcentrationfromthegraphforthispointfromthelogaxis.Thisistheisopotentialvalue.
