Sample preparation
• Mycophenolic acid and its glucoronide are typically measuredin plasma since they are not found in any other blood fractionanalysedanalysed
• However, Cyclosporine A, everolimus, sirolimus andtacrolimus are found distributed throughout blood fractionsgand must be measured in whole blood

Sample Preparation – Protein Crash + ZnSO4
100 µL of plasma or whole blood+
200 µL of precipitating reagent (1:4 v/v 0.4M ZnSO4:methanol) containing internal standards
Vortex for 30 seconds
Centrifuge @ 10000 rpm for 4 minutes
Transfer clear supernatant in autosampler vialp p

MPA-G is susceptible to in-source fragmentation where glucuronide is easily lost.
